摘 要:研究绿豆皮牡荆素对过氧化氢(H2O2)损伤人脐静脉内皮细胞(HUVEC)的保护作用。H2O2诱导建立HUVEC细胞损伤模型,设置空白组、绿豆皮牡荆素低、中、高剂量组、VC对照组,噻唑蓝和试剂盒法分别测定细胞存活率、目标成分(MDA、GSH、SOD、LDH、GSH-Px)。最佳H2O2损伤浓度为900μmol/L此时细胞存活率为51.32%;剂量组质量浓度为60、80、100μg/mL时,细胞存活率分别为110.61%、122.30%、131.68%,不同剂量组均能显著降低细胞及培养液中MDA含量,增加GSH含量,提高SOD、GSH-Px活性;能显著提高细胞中LDH活性,降低培养液中LDH活性;绿豆皮牡荆素质量浓度为100μg/mL时,细胞保护能力与同浓度下VC相当。绿豆皮牡荆素能促进HUVEC细胞增殖,抑制自由基生成,增强抗氧化酶活性,降低细胞氧化损伤程度,且存在剂量依赖关系。The aim of this study was to investigate the protective effect of vitexin from mung bean skin(MBS)on human umbilical vein endothelial cells(HUVEC)damaged by hydrogen peroxide(H2O2).The HUVEC cell damage model induced by H2O2 was established by.and the experiments were divided into blank,low-,medium-and high-dose MBS,VC control group.The cell survival rate and target components(MDA、GSH、SOD、LDH、GSH-Px)were determined by the method of methyl thiazolyl tetrazolium(MTT)and the kit method.The optimal concentration of H2O2 was 900μmol/L,and the cell survival rate was 51.32%.When the concentration of the dose group was 60,80 and 100μg/mL,the cell survival rates were 110.61%,122.30%,and 131.68%,respectively.In all dose groups of vitexin from MBS,the content of MDA was significantly decreased in cells and culture medium,however,the content of GSH was significantly increased and the activities of SOD,GSH-Px were significantly increased.The LDH activity was significantly increased in cells and significantly reduced in culture medium.The cell protection ability was significantly equivalent to VC when the drug concentration was 100μg/mL.The vitexin from MBS can promote he proliferation of HUVEC cells,inhibit the generation of free radicals,enhance the activity of antioxidant enzymes,and reduce the degree of cell oxidative damage in a dose-dependent manner.
