摘  要:【目的】研究甘肃14种蜂蜜黄酮含量及黄酮提取液的抑菌性能差异.【方法】以甘肃成县土蜂蜜为原料,通过单因素和响应面设计优化树脂静态吸附提取黄酮的工艺参数,以优化后的参数提取并测定14种蜂蜜的黄酮含量,并用牛津杯法比较分析其抑菌性能.【结果】蜂蜜黄酮提取优化参数为蜂蜜20 g,pH=1.78盐酸吸附液200 mL、XAD-4树脂添加量为23.88 g、92.2%乙醇解析液100 mL;验证试验蜂蜜黄酮提取量为0.0750 mg/g.14种甘肃蜂蜜黄酮含量最高为土蜂蜜0.0750 mg/g,最低为板蓝根蜜0.0136 mg/g.抑菌试验结合SPSS显著性分析显示荞麦蜜黄酮提取液对金黄色葡萄球菌、油菜蜜黄酮提取液对单增李斯特氏菌和铜绿假单胞菌、板蓝根蜜黄酮提取液对藤黄微球菌的抑菌率分别达到了对照青霉素G标准溶液的60.55%、38.77%、70.52%和44.74%.【结论】14种蜂蜜黄酮提取液都具有抑菌性,黄酮含量较低的板蓝根蜜黄酮提取液比土蜂蜜具有更好的抑菌性,可能与蜜源植物中存在的抑菌物质有关,蜂蜜抑菌机理仍待进一步研究.本研究结果为蜂蜜总黄酮提取及其抑菌应用提供了依据.【Objective】In order to study the content of flavones and the antibacterial activity of the extract substance in 14 kinds of Gansu honey.【Method】The wild honey was collected from Chengxian of Gansu,the technological parameters of the static adsorption extracting flavones with resin was optimized by single factor and response surface design,the content of flavones was compared,and the antibacterial activity was assayed with Oxford Cup.【Result】The optimum extraction parameters of flavones from honey were as follows:honey 20 g,pH 1.78 hydrochloric acid adsorption solution 200 mL,the XAD-4 resin 23.88 g,92.2%ethanol solution 100 mL.The honey flavones extracted content of verification test was 0.0750 mg/g.The content of flavones of the wild honey was highest 0.0750 mg/g and the content of flavones of Radix isatidis honey was the lowest 0.0136 mg/g.The antibacterial inhibition rate was 60.55%,38.77%,70.52%and 44.74%of buckwheat honey flavones against Staphylococcus aureus,rape honey flavones against Listeria monocytogenes,Pseudomonas aeruginosa and Radix Isatidis flavones against Micrococcus luteus when compared with the control penicillin G standard solution,respectively.【Conclusion】The flavones extraction from 14 kinds of honey all has antibacterial activity,and the Radix isatidis honey with lower flavones content,but it's flavones extraction had better antibacterial activity than wild honey,which might be related to the antibacterial substances existing in the nectar plants.
