摘 要:该文以蚕蛹粉和麸皮为原料利用全细胞发酵制备蚕蛹多肽,并对制备蚕蛹多肽的各项参数进行优化,以提高水解后多肽得率。通过正交试验分析,得到最佳工艺条件为:水解温度30℃、水解时间3 d、加酶量2368 U/g。在最优条件下水解蚕蛹蛋白,多肽得率21.35 mg/mL,水解度4.58%。通过方差分析可知,水解温度、水解时间均对蚕蛹多肽得率和水解度有显著性影响,而加酶量影响不显著。用SephadexG-25凝胶树脂对多肽进行洗脱得到两个不同组分的肽段,并通过抗氧化性研究得到组分Ⅰ和组分Ⅱ对·OH具有很强的清除能力,均达到90%以上。Using silkworm pupa meal and bran as raw materials,silkworm pupa polypeptide was produced by solid-state fermentation of whole cells.The parameters of polypeptide production were optimized to maximize its post-hydrolysis yield.By analysis of orthogonal experiments,the optimal parameters for polypeptide preparation were found to be:hydrolysis temperature of 30℃,hydrolysis time of 3 d,and enzyme activity of 2368 U/g.The polypeptide yield was 21.35 mg/mL,and the hydrolysis rate was 4.58%using the optimized hydrolysis process.Analysis of variance indicated that hydrolysis temperature and time each had significant effects on polypeptide yield and degree of silkworm pupa meal hydrolysis,while enzyme volume had no significant effect.Using Sephadex G-25 gel filtration chromatography to isolate polypeptide,component I and component II were obtained.Both components display·OH scavenging capacity,with scavenging rates of>90%.
