摘  要:作为药食同源的青刺果因具有抗氧化、提高免疫力等生物活性已被人们长期食用,但建立一种操作简单、提取率高且提取物生物活性强的提取方法仍是青刺果当前亟需解决的问题。该研究利用水酶法结合超声波技术处理胶体磨制得的青刺果样液,在单因素实验的基础上结合响应面法优化提取工艺,并对提取物进行抗氧化活性评价及成分分析。结果表明,采用5%的青刺果溶液加入2 g的中性蛋白酶,对青刺果中抗氧化活性成分的提取效果较好。利用响应面法优化后,当水酶法的酶解温度为57℃、超声时间为26 s、功率为120 W时得到的提取物对DPPH·的清除率可达85.88%±0.15%,比理论预测值仅低了0.33%。通过气相色谱-质谱法(GC-MS)分析表明,提取液中含有油酸酰胺和棕榈酰胺等不饱和脂肪酰胺类物质成分为69.44%。该研究建立的提取工艺能获得含量较高的青刺果抗氧化活性成分,可为高原特色植物的进一步开发利用提供理论基础。As a homology of medicine and food,Prinsepia utilis Royle has been consumed by people for a long time because of its biological activity such as antioxidant and immunity enhancement.However,it is still an urgent problem to establish an extraction method with simple operation,high extraction rate and strong biological activity.In this study,the aqueous enzymatic method combined with ultrasonic technology is used to treat Prinsepia utilis Royle solution made by colloid grinder.The extraction process is optimized by response surface methodology(RSM)based on single factor test,and the antioxidant activity of the extract is evaluated and its components are analyzed.The results show that adding 2 g neutral protease in 5%solution of Prinsepia utilis Royle has better extraction effect of antioxidative active components.The DPPH·scavenging rate of the extract is 85.88%±0.15%,which is only 0.33%lower than the predicted value when the hydrolysis temperature is 57℃,ultrasonic time is 26 s and power is 120 W.Gas chromatography-mass spectrometry(GC-MS)analysis results show that the content of unsaturated fat amides such as oleate amide and palmitamide is 69.44%.The extraction process established in this study can obtain higher content of antioxidative active components of Prinsepia utilis Royle,which can provide a theoretical basis for further development and utilization of plateau characteristic plants.
