摘  要:耐高温的细菌是中高温大曲的重要优势菌,为了鉴定特香型白酒大曲的功能细菌,从特香型大曲火圈层中分离筛选到1株具有水解淀粉活性的耐高温细菌菌株ZRF-1。本试验结合扫描电镜鉴定菌株形态、生理生化特性鉴定和gyrB基因序列同源性分析鉴定该菌株为地衣芽孢杆菌。利用该菌株模拟大曲制作工艺固态发酵产生吡嗪类风味化合物四甲基吡嗪、2,3,5-三甲基吡嗪和2,5-二甲基吡嗪,以及大量的乙偶姻。ZRF-1菌株在产酶培养基中摇瓶发酵的淀粉酶活力可达53.42 U/mL。初步确定地衣芽孢杆菌ZRF-1菌株为特香型大曲的功能菌株,可以丰富白酒酿造的菌种资源库。Heat-resistant bacteria are important dominant bacteria in medium and high temperature Daqu.In order to identify the func-tional bacteria of Texiang Daqu,a heat-resistant strain ZRF-1 with starch hydrolysis activity was isolated from the fire circle layer of Texiang Daqu.By using scanning electron microscopy,physiological and biochemical experiments,and gyrB gene sequence homolo-gy analysis,the strain was identified as Bacillus licheniformis.The strain was used in the simulated solid-state fermentation process of Daqu,and pyrazine flavor compounds,including tetramethylpyrazine,2,3,5-trimethylpyrazine and 2,5-dimethylpyrazine,and large amounts of acetoin were produced.The amylase activity of strain ZRF-1 was 53.42 U/mL in enzyme-producing medium by shake flask culture.It is preliminarily determined that the Bacillus licheniformis strain ZRF-1 is a functional strain of Texiang Daqu,which expands the strain resource repository of Baijiu production.
