摘 要:根据前研究结果及实验室条件,以壳聚糖为载体,戊二醛为交联剂固定化复合酶,从粉碎过的革胡子鲶下脚料匀浆中提取磷脂酰胆碱,考察载体形状、固定化复合酶加酶量、固定化酶酶解时间及酶解pH对磷脂酰胆碱提取率的影响。单因素试验结果表明:最佳酶固定化载体形状为田字形,最佳固定化复合酶加酶量为0.05%,最佳固定化酶解时间为120 min,最佳固定化酶解pH为5。固定化酶重复使用7次后,提取率仍然保持在50%以上。在三因素三水平的正交试验中,最佳组合为固定化复合酶添加量0.05%、酶解时间90 min、酶解pH 5。该提取工艺可靠,并且参数稳定,为提取鱼下脚料中的磷脂酰胆碱提供了新的思路与方法。According to previous research results and laboratory conditions,phosphatidylcholine was extracted from crushed catfish offal homogenate by immobilized complex enzyme with chitosan as carrier and glutaraldehyde as crosslinking agent.The effects of carrier shape,amount of immobilized complex enzyme,hydrolysis time and pH of immobilized enzyme on the extraction rate of phosphatidylcholine were investigated.The results of single factor experiment showed that the best carrier shape was Tian shape,the best amount of immobilized compound enzyme was 0.05%,the best immobilized enzymatic hydrolysis time was 120 min,and the best pH of immobilized enzymatic hydrolysis was 5.After the immobilized enzyme was reused for 7 times,the extraction rate remained above 50%.In the orthogonal experiment of three factors and three levels,the best combination was 0.05%of immobilized compound enzyme,enzymatic hydrolysis time 90 min and enzymatic hydrolysis pH 5.The extraction process was reliable and the parameters were stable,which provided a new idea and method for extracting phosphatidylcholine from fish residues.
