摘 要:以何首乌叶为原料,分别用石油醚、二氯甲烷、乙酸乙酯、正丁醇、水对其乙醇提取物进行萃取,研究其不同萃取部位的总黄酮及总多酚含量,通过ABTS、DPPH、Fe^(3+)还原能力测定等方法综合评估其不同萃取相的抗氧化能力,并运用Pearson法分析总黄酮及总酚的含量与其抗氧化活性之间的相关性。结果表明,何首乌叶乙酸乙酯萃取部位的总黄酮含量与总酚含量最高,分别为415.70±8.97 mg/g和257.44±1.32 mg/g。各萃取相抗氧化活性为乙酸乙酯部位>粗提物>正丁醇部位>石油醚部位>二氯甲烷部位>水部位。各部位抗氧化活性与其总黄酮、总多酚含量呈显著正相关(p<0.05),黄酮类及酚类化合物可能是发挥抗氧化作用的主要物质,为综合开发利用何首乌叶资源提供理论基础。The ethanol extract of Polygonum multiflorum Thunb.leaves was extracted with petroleum ether,dichloromethane,ethyl acetate,n-butanol and water,respectively,and the total flavonoid and phenolic content of different extract fractions were studied.ABTS,DPPH and ferric reducing antioxidant power methods were used to evaluate the antioxidant activity of different fractions comprehensively.The correlation between the contents of total flavonoids and total polyphenols and their antioxidant activity were analyzed by Pearson method.The results suggested that the total flavonoid and phenolic contents of ethyl acetate fraction of Polygonum multiflorum leaves were highest and the contents were 415.70±8.97 mg/g and 257.44±1.32 mg/g,respectively.The antioxidant activity of each fraction was as follows:ethyl acetate fraction>crude extract>n-butanol fraction>petroleum ether fraction>dichloromethane fraction>water fraction.The antioxidant activity of each fraction was positively correlated with the total flavonoid and phenolic content(p<0.05).It was indicated that the flavonoids and phenolic compounds might be the main substances of antioxidation.A theoretical basis for the comprehensive development and utilization of Polygonum multiflorum resources was provided.
