摘  要:选用油莎豆粕为原料,以蛋白质提取率为指标,采用纤维素酶辅助碱法提取油莎豆粕中的蛋白质。在单因素试验的基础上,运用Box-Benhnken响应面法优化纤维素酶辅助碱法提取油莎豆粕蛋白的最优工艺条件,并与碱提酸沉法进行对比。结果表明:最佳提取工艺为料液比1∶21.7(g/mL)、加酶量0.10%(以油莎豆粕干粉质量计)、酶解温度42.1℃、酶解pH 4.4、酶解时间1.0 h,在此条件下油莎豆粕蛋白提取率高达71.91%,远高于碱提酸沉法,此工艺很大程度上提高了油莎豆粕蛋白提取率。Cyperus esculentus L.meal was selected as the raw material,and the protein extraction rate was used as the index,and the protein in the Cyperus esculentus L.meal was extracted by cellulase-assisted alkaline method.On the basis of single factor experiment,Box-Benhnken response surface methodology was used to optimize the optimal process conditions of cellulase-assisted alkaline extraction of Cyperus esculentus L.meal protein,and compared with alkaline extraction and acid precipitation method.The results showed that the optimal extraction process was as follows:the ratio of solid to liquid 1∶21.7(g/mL),the amount of enzyme added 0.10%(calculated on the basis of the dry powder of Cyperus esculentus L.meal),the enzymatic hydrolysis temperature 42.1℃,the enzymatic hydrolysis pH 4.4,and the enzymatic hydrolysis time 1.0 h.Under this condition,the extraction rate of Cyperus esculentus L.meal protein was as high as 71.91%,which was much higher than that of the alkaline extraction and acid precipitation method.The process greatly improved the extraction rate of Cyperus esculentus L.meal protein.
