摘 要:目的 通过为噬菌体添加纳米涂层,探究其对噬菌体热稳定性的影响。方法 模拟自然界微生物的生物矿化方式,利用偏硅酸钠为噬菌体添加一层纳米涂层。分析了不同浓度偏硅酸钠对噬菌体矿化的影响。同时通过体外浊度法检测了矿化对噬菌体感染能力以及对热稳定性的影响。结果 高浓度的偏硅酸钠有利于提高噬菌体的负载率,在浓度为100mmol/L时负载率为99.2%。与未经矿化的噬菌体(24h时,OD为1.0)相比,矿化不影响副溶血性弧菌噬菌体(24 h时, OD为0.83~1.17)对宿主细菌的感染能力。矿化提高了噬菌体对温度的抗性,矿化前噬菌体在80℃下处理30 min后失去感染能力,但矿化噬菌体在90℃下处理120 min后仍然具有和原始噬菌体相似的感染能力。结论 该方法能够在不影响副溶血性弧菌噬菌体对细菌抑制效果的情况下提高噬菌体的耐热性。Objective To explore its influences on the thermal stability of phage by adding nano-coating to phage.Methods In order to simulate the biomineralization of microorganisms in nature,sodium metasilicate was used to add a layer of nano-coating to the phage.The effect of different concentrations of sodium metasilicate on the mineralization of phage was analyzed.At the same time,the effect of mineralization on phage infection ability and thermal stability was detected by in vitro turbidity method.Results High concentration of sodium metasilicate was conducive to improve the loading rate of phage,and the loading rate was 99.2% at 100 mmol/L.The ability of host bacteria to be infected by Vibrio parahaemolyticus phage(ODat 24 h was 0.83 to 1.17) was not affected by mineralization,as compared to unmineralized phage(ODat 24 h was 1.0).The resistance of phage to temperature was improved by mineralization,the phage before mineralization lost the infectious ability after being treated at 80℃for 30 min,but the mineralized phage still had the similar infectious ability to the original phage after being treated at 90℃ for 120 min.Conclusion The method can improve the heat resistance of Vibrio parahaemolyticus phage without affecting the bacteriostasis effect of phage.
