摘 要:通过单因素试验和正交试验确定了一种离子液体双水相体系的最佳萃取条件,与高效液相色谱(UPLC)技术相结合,建立了一种检测食醋中杂环胺(HAAs)类物质Harman和Norharmane的方法。结果表明,最佳萃取条件为选用[C_(6)MIM]Br-K_(2)HPO_(4)离子液体双水相体系,[C_(6)MIM]Br用量200μL,K_(2)HPO_(4)用量0.5 g,pH值10,超声时间3 min;在此条件下,Harman和Norharmane的线性范围均为10.0~400.0μg/mL,相关系数R^(2)均为0.9995,LODs分别为0.50μg/mL和0.45μg/mL,精密度、重复性和回收率均良好,即该方法适用于检测食醋中的Harman和Norharmane。A method of ionic liquid aqueous two-phase system,in which optimum extraction conditions were achieved by both single factor experiment and orthogonal experiment,combined with ultra-high performance liquid chromatography(UPLC),was established to detect Harman and Norharmane in vinegar.The experimental results showed that the optimum extraction conditions were as follows:[C_(6)MIM]Br-K_(2)HPO_(4) were used as the ionic liquid aqueous two-phase system,the amount of[C_(6)MIM]Br was 200μL,the amount of K_(2)HPO_(4) was 0.5 g,pH value was 10,and the ultrasound time was 3 min.Under the appropriate optimal conditions,both the linear equations of Harman and Norharmane were 10.0~400.0μg/mL,and both the correlation coefficients R^(2) were 0.9995.The limits of detection(LODs)of Harman and Norharmane were 0.50μg/mL and 0.45μg/mL,respectively.All the precision experiment,repeatability experiment,and recovery experiment showed that the method had better accuracy and precision.That is,the method could be used for the determination of Harman and Norharmane in vinegar.
