摘 要:为评价野梧桐种子的生物活性,采用Folin-Ciocalteu比色法测定野梧桐种子石油醚提取物(Ⅰ)和95%乙醇提取物(Ⅱ)及其乙酸乙酯萃取物(Ⅲ)和正丁醇萃取物(Ⅳ)的总酚含量;采用体外活性测试方法,分别对其α-淀粉酶、α-葡萄糖苷酶和醛糖还原酶抑制活性及抗氧化活性(DPPH、ABTS和FRAP法)进行评价。结果表明:乙酸乙酯萃取物总酚含量最高,达到(679.84±14.65)mmol·g^(-1);除石油醚提取物(Ⅰ)外,Ⅱ、Ⅲ和Ⅳ均具有一定的酶抑制活性及抗氧化活性,其中乙酸乙酯萃取物(Ⅲ)活性最为显著,对α-淀粉酶、α-葡萄糖苷酶和醛糖还原酶的半数抑制浓度(IC_(50))分别为0.106、0.087、0.058 mg·mL^(-1),清除DPPH、ABTS自由基的IC分别为0.047、0.031 mg·mL^(-1),FRAP值为12.118 mmol·g^(-1)。这说明野梧桐种子可应用于Ⅱ型糖尿病及其慢性并发症的防治,酚类成分可能是其主要活性成分。Inhibiting the activities ofα-amylase,α-glucosidase and aldose reductase and suppressing oxidative stress are the an effective means to prevent and treat type 2 diabetes(T2 D)and its related chronic complications.The total phenolic contents of petroleum ether,95%ethanol,ethyl acetate and n-butanol extracts(Ⅰ—Ⅳ)from Mallotus japonicus seeds were determined by Folin-Ciocalteu assay.In addition,theα-amylase,α-glucosidase,aldose reductase inhibitory activities as well as the antioxidant activity(DPPH,ABTS,FRAP)of fractionsⅠ—Ⅳwere evaluated in vitro.The result showed that the ethyl acetate fraction(Ⅲ)possessed the highest total phenolic content of(679.84±14.65)mmol·g^(-1).Moreover,ethyl acetate fraction(Ⅲ)exhibited the strongest inhibitory activities onα-amylase,α-glucosidase and aldose reductase with ICvalues of 0.106,0.087,0.058 mg·mL^(-1),respectively,and the most significant antioxidant activities with ICvalues of 0.047 mg·mL^(-1)(DPPH),0.031 mg·mL^(-1)(ABTS)were observed,while FRAP value was 12.118 mmol·g^(-1).These findings suggested that polyphenols might be the main active constituents of M.japonicas seeds for preventing and treating T2 D and its related chronic complications.
