摘 要:Micro RNA是一种由20~24个核苷酸组成的短链非编码RNA,可以通过与靶基因3’UTR结合调控基因表达,但miR-214在奶牛泌乳过程中的研究还很少。本研究应用奶牛乳腺上皮细胞模型,通过过表达miR-214,检测奶牛乳腺上皮细胞中乳糖、乳脂和乳蛋白的合成情况。结果表明,在奶牛乳腺上皮细胞内转染miR-214后,miR-214的表达显著上升,而乳糖、乳脂和乳蛋白的含量均显著下降,说明miR-214能够抑制乳糖、乳脂和乳蛋白的合成,影响奶牛乳腺上皮细胞的泌乳功能。Micro RNA is a short non-coding RNA composed of 20-24 nucleotides, which can regulate gene expression by binding to the3’UTR of the target gene. However, there are few studies on miR-214 during lactation in dairy cows. In this study, a cow mammary epithelial cell model was used to detect the synthesis of lactose, milk fat and milk protein in cow mammary epithelial cells by overexpressing miR-214. The results showed that after transfection of miR-214 in cow mammary epithelial cells, the expression of miR-214 was significantly increased, while the contents of lactose, milk fat and milk protein were significantly decreased, indicating that miR-214 can inhibit the synthesis of milk lactose, milk fat and milk protein, and affect the lactation function of bovine mammary epithelial cells.
