摘  要:该文通过采集不同生境的土样,筛选得到了能利用果胶生物质或其产物产油脂的两株酵母菌2S-6和HN-3B,对应油脂含量分别为51.44%和64.66%,其中菌株2S-6通过分子鉴定为奥默柯达菌,且该菌还具有优良的分解果胶能力,表明其具有较强活性的果胶酶,可用于果胶降解酶膜反应器的构建。因此,通过基因工程技术,获得其大量非糖基化多聚半乳糖醛酸酶之后,在1.25%戊二醛交联处理2.55 h的最佳条件下,将其固定于静电纺丝壳聚糖纳米纤维膜上制得了酶膜反应器,并将果胶水解为半乳糖醛酸单体,转化率可达到63.96%。这为实现果胶生物质废料的资源化利用和减少环境污染奠定基础。This paper discusses two strains of yeast,2S-6 and HN-3B,which are screened from collected soil samples from different habitats.2S-6 and HN-3B can produce oil by using pectin biomass or its products,corresponding to the oil content of 51.44% and 64.66%,respectively.The 2S-6 strain has been identified as Kodamaea ohmeri through molecular identification,and it also has excellent ability to decompose pectin,indicating that it has strong pectinase activity and can be used for the construction of enzyme membrane reactor for pectin degradation.Therefore,a large amount of non-glycosylated polygalacturonase was obtained through genetic engineering technology.Under the optimal conditions of 2.55 hours cross-linking treatment with 1.25% glutaraldehyde,it was fixed on the chitosan nanofiber membrane of electrospinning to prepare the enzyme membrane reactor,and pectin was hydrolyzed into galacturonic acid monomer with a conversion rate of 63.96%.This will lay a foundation for realizing the resource utilization of pectin biomass waste and reducing environmental pollution.
