摘  要:目的:研究椰毒假单胞菌酵米面亚种在培养基中的生长规律和产毒情况,为椰毒假单胞菌酵米面亚种的菌株特性研究提供参考。方法:接种不同初始菌液的马铃薯葡萄糖水(PD水),静置培养72 h,测定不同培养温度下的菌株和米酵菌酸浓度;接种同一浓度菌液的PD水,测定其不同培养方式下的米酵菌酸浓度。结果:静置培养后,低、中、高浓度菌液在4℃条件下72 h内未出现明显繁殖,但在26℃、36℃条件下培养出现明显繁殖,达到10^(8) CFU·mL^(-1)数量级后趋于平稳;4℃条件下的低、中、高浓度菌液和26℃的低、中浓度菌液培养物中均未检出米酵菌酸,26℃培养条件下的高浓度菌液培养物中检出的米酵菌酸浓度较低,为5.48~6.72μg·kg^(-1)。而36℃条件下的3个浓度菌液培养物中的米酵菌酸持续升高,但都低于400μg·kg^(-1)。接种了10^(8) CFU·mL^(-1)菌液的PD水采用静置、振荡培养后,米酵菌酸浓度振荡培养>静置培养。结论:椰毒假单胞菌酵米面亚种在PD水中培养,其菌液浓度和米酵菌酸的产量与培养温度呈正相关,36℃和振荡方式培养有利于米酵菌酸的产生。Objective:To provide a reference for the study of the strain characteristics of Pseudomonas cocovenenans subsp.farinofermentans,the growth rules and toxin production of Pseudomonas cocovenenans subsp.farinofermentans in the culture medium were analyzed.Method:The potato glucose water(PD water)was inoculated with different initial bacterial fluids,resting incubated for 72 h to determine the concentration of bacterial strain and bongkrekic acid at different culture temperatures.The PD water was inoculated with the same concentration of bacteria fluids,and determined concentration of bongkrekic acid under different culture modes.Result:After static culture,the low,medium and high concentrations of bacterial solution did not reproduce significantly within 72 hours at 4℃,but did reproduce significantly at 26℃and 36℃,reaching 108 CFU·mL-1;No bongkrekic acid was detected in the low,medium and high concentrations of bacterial fluid at 4℃and the low and medium concentrations of bacterial fluid culture at 26℃.The concentration of bongkrekic acid detected in the high concentrations of bacterial fluid culture at 26℃was low,5.48~6.72μg·k g^(-1).At 36℃,the rice yeast acid in the three concentrations of bacterial liquid culture continued to increase,but all were lower than 400μg·kg^(-1).After the PD water inoculated with 108 CFU·mL^(-1) bacterial solution was incubated by standing and shaking,the concentration of rice yeast acid in shaking culture was higher than that in standing culture.Conclusion:Pseudomonas cocovenenans subsp.farinofermentans was cultured in PD water,and its bacterial fluid concentration and the bongkrekic acid was positively correlated with the culture temperatures,and the culture at 36℃and shaking mode was conducive to the production of bongkrekic acid.
