摘 要:采用超声破碎复合纤维素酶酶解的方法优化陈皮中精油的提取工艺,并测定用此方法提取的陈皮精油对DPPH·(1,1-二苯基-2-三硝基苯肼)和ABTS^(+)·(2,2′-联氮-二(3-乙基-苯并噻唑-6-硫磺))清除率的影响。研究表明,影响陈皮精油产率的主次顺序为纤维素酶添加量>超声功率>酶解时间>酶解温度>超声时间。正交试验结果表明,纤维素酶的添加量4%、酶解温度45℃、酶解时间120 min、超声功率80%(640 W)、超声时间40 min为陈皮精油的最佳提取条件,在此条件下陈皮精油的产率为0.69%。抗氧化试验表明,浓度高于40 mg/mL的陈皮精油对DPPH·有较好的清除能力,浓度高于10 mg/mL的陈皮精油对ABTS^(+)·有较好的清除能力。通过超声破碎复合纤维素酶酶解的方法优化陈皮中精油的提取工艺,可以促进柑橘果皮的综合利用,提高柑橘产业的附加值,具有良好的利用前景。The extraction process of essential oil from Citri reticulatae pericarpium(CRP)is optimized by ultrasonication combined with cellulase hydrolysis,and the effects of CRP essential oil extracted by this method on the scavenging rate of DPPH·(1,1-diphenyl-2-trinitrophenyl hydrazine)and ABTS^(+)·(2,2'-diazo-di(3-ethyl-benzothiazole-6-sulfur))are determined.The research shows that the primary and secondary order affecting the yield of CRP essential oil is cellulase addition amount>ultrasonic power>enzymatic hydrolysis time>enzymatic hydrolysis temperature>ultrasonic time.The orthogonal experimental results show that the optimum extraction conditions of CRP essential oil are as follows:the addition amount of cellulase is 4%,the enzymatic hydrolysis temperature is 45℃,the enzymatic hydrolysis time is 120 min,the ultrasonic power is 80%(640 W)and the ultrasonic time is 40 min.Under these conditions,the yield of CRP essential oil is 0.69%.The antioxidation experiment shows that CRP essential oil with the concentration higher than 40 mg/mL has good scavenging ability on DPPH·and CRP essential oil with the concentration higher than 10 mg/mL has good scavenging ability on ABTS^(+)·.Optimizing the extraction process of essential oil from CRP by ultrasonication combined with cellulase hydrolysis can promote the comprehensive utilization of citrus peel and improve the added value of citrus industry,which has a good utilization prospect.
