摘 要:为探究黄精速溶茶酶解最佳工艺参数,以黄精速溶茶得率为响应值,以液料比、加酶量、酶解温度和酶解时间4个因素为自变量,通过响应面法进行优化,得到最佳工艺条件。结果表明:黄精速溶茶的最佳工艺条件为液料比21∶1(mL/g)、加酶量1.4%、酶解温度47℃、酶解时间37 min,在此条件下,黄精速溶茶得率为26.08%,与响应面预测值26.82%相近;超声复合酶法浸提的黄精速溶茶色泽呈黄色,黄精多糖含量为3.73%,黄酮含量为0.95%;抗氧化试验表明,黄精速溶茶对1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基和超氧阴离子自由基的IC50分别为14.34 mg/mL和16.45 mg/mL,且其对自由基的清除能力随着其浓度的增加而增大。The parameters of enzymatic hydrolysis were explored for the production of Polygonatum sibiricum instant tea.With the yield of P.sibiricum instant tea as the response value and the liquid-to-material ratio,enzyme dosage,enzymatic hydrolysis temperature and time as the independent variables,the processing parameters of the instant tea were optimized by response surface methodology.The results showed that the optimum enzymatic hydrolysis parameters were liquid-to-material ratio of 21∶1(mL/g),enzyme dosage of 1.4%,and enzymatic hydrolysis at 47℃for 37 min.Under these conditions,the yield of P.sibiricum instant tea was 26.08%,which was close to the predicted value of 26.82%.The P.sibiricum instant tea extracted by ultrasound combined with compound enzyme was yellow,with the polysaccharide content of 3.73%and the flavonoid content of 0.95%.The IC50 values of Polygonatum sibiricum instant tea for scavenging 1,1-diphenyl-2-picrylhydrazyl(DPPH)free radicals and superoxide free radicals were 14.34 mg/mL and 16.45 mg/mL,respectively.Moreover,the free radical scavenging ability of the instant tea increased with the increase in the concentration.
