摘  要:为探讨紫色杆菌素(violacein,Vio)的稳定性和生物活性,该文研究紫色杆菌素的热稳定性、pH值稳定性、抑菌和抗氧化活性,并将其内化于聚乙烯醇(polyvinyl alcohol,PVA)膜液,制备PVA/Vio膜,分析PVA/Vio膜包裹对草鱼保鲜效果和货架期的影响。结果表明,紫色杆菌素在pH1~10、0℃~80℃条件下性质稳定,对金黄色葡萄球菌、化脓性链球菌、单增李斯特菌和蜡样芽孢杆菌的最小抑菌浓度分别为3.13、6.25、12.50、6.25 mg/L,对ABTS^(+)自由基和DPPH自由基清除能力的IC_(50)分别为1.64 mg/L和14.72 mg/L;当浓度为12.50 mg/L时,紫色杆菌素的总还原能力为88.49μmol/L;PVA/Vio膜具有良好的抑菌和抗氧化活性,4℃冷藏条件下用PVA/Vio膜包裹的草鱼菌落总数和硫代巴比妥酸反应物值均大幅低于PVA膜和普通的聚乙烯保鲜膜,其货架期延长4 d左右,保鲜效果十分明显。The thermostability,pH stability,and antibacterial and antioxidant activities of violacein were determined.Further,violacein was used to prepare the polyvinyl alcohol/violacein(PVA/Vio)composite film with the solution of PVA.The film was then used to preserve grass carp and the preservation effect and shelf-life were evaluated.The results showed that violacein had stable activity within the ranges of pH1-10 and 0℃-80℃.The minimum inhibitory concentrations of violacein for Staphylococcus aureus,Streptococcus pyogenes,Listeria monocytogenes,and Bacillus cereus were 3.13,6.25,12.50 mg/L and 6.25 mg/L,respectively.The IC_(50) of violacein for scavenging ABTS^(+)and DPPH free radicals were 1.64 mg/L and 14.72 mg/L,respectively.The violacein of 12.50 mg/L showed the total reduction ability of 88.49μmol/L.In addition,the PVA/Vio film has good antioxidant and antibacterial activities.Compared with PVA film and polyethylene film,the PVA/Vio film improved the preservation effect on the grass carp stored at 4℃,inhibited the increases in total viable count and thiobarbituric acid reactive substances,and prolonged the shelf-life by about 4 days.
