摘  要:以孔鳐加工的副产物软骨为原料,利用响应面方法优化鳐鱼硫酸软骨素提纯工艺,并对获得的鳐鱼硫酸软骨素进行质量及结构的表征。结果表明,最优提取工艺为碱液浓度2.3%、提取温度43℃、酶添加量0.7%时,硫酸软骨素提取率为43.32%。提取的鳐鱼硫酸软骨素符合国药标准,其重均分子量、数均分子量和多分散系数分别为40 752、38 257和1.065 2,经红外光谱分析,鳐鱼硫酸软骨素与鲨鱼硫酸软骨素具有相同结构组织,均由β-1,3糖苷键连接的β-D-葡萄糖醛酸和N-乙酰基-β-D-氨基半乳糖组成,该研究结果不仅可大幅度增加鳐鱼加工产业的附加值,也可为硫酸软骨素的工业生产提供一定的技术支持。In this work,the cartilage of Raja porosa was employed as a raw material,the extraction process was optimized by the response surface method,and the quality and structure of the obtained chondroitin sulfate were characterized.The results showed that the optimal conditions for maximal extract yield(43.32%)were 2.3% of NaOH,43 ℃ of extraction temperature,and 0.7% of alkaline protease.The extracted substance met the national drug standard.The average molecular weight,number average molecular weight,and polydispersity coefficient were 40 752,38 257,and 1.065 2,respectively.The structure of chondroitin sulfate from R.porosa was the same as that of sharks,which was made up of β-D-glucuronic acid and N-acetyl-β-D-galactosamine connected by β-1,3 glucoside bond.This study has increased the added value of R.porosa processing industry and provided technical support for the industrial production of chondroitin sulfate in the future.
