摘  要:为探究4种复合生物保鲜剂对冰鲜菊黄东方鲀的作用效果,测定了菌落总数并采用Illumina Miseq高通量测序方法对复合生物保鲜剂处理的菊黄东方鲀肌肉细菌的16S rRNA基因的2个高变区(V3~V4)进行测序分析。结果显示,复合生物保鲜剂A(0.5%茶多酚+0.2%乳酸链球菌素+0.3%溶菌酶)可使菊黄东方鲀贮藏期间的菌落总数显著降低(P<0.05)。冰鲜18 d时的细菌群落多样性最高,表明复合生物保鲜剂A可将菊黄东方鲀冰鲜保存时间延长到18 d或以上。复合生物保鲜剂B(0.5%茶多酚+0.2%乳酸链球菌素)、C(0.2%乳酸链球菌素+0.3%溶菌酶)和D(0.5%茶多酚+0.3%溶菌酶)可将菊黄东方鲀冰鲜保存时间货架期延长至15 d左右。冰鲜条件下菊黄东方鲀肌肉5组15个样品细菌分布于21门187属,优势菌门分别为变形菌门、放线菌门和厚壁菌门,主要优势菌属包括假单胞菌属、无色杆菌属与红球菌属。复合生物保鲜剂改变了菊黄东方鲀肌肉的群落结构,菊黄东方鲀特定腐败菌可能为假单胞菌属。试验结果为后期开展菊黄东方鲀保鲜技术研究提供了理论参考。The effects of compound biological preservative on microbial community diversity of Takifugu flavidus during chilled storage were investigated.The aerobic plate count was measured and Illumina MiSeq high-throughput sequencing method was used to analyze the two high-variation regions(V3-V4)of 16S rRNA gene of microbial structure of the T.flavidus muscle treated with the compound biological preservatives.The results showed that compound biopreservative A(0.5% tea polyphenol+0.2% nisin+0.3% lysozyme)significantly reduced the aerobic plate count(P<0.05).The diversity of the bacterial community was the highest at the18th day,which indicated that compound biological preservative A could prolong the preservation time of fresh T.flavidus to 18 days or more.Compound biopreservatives B(0.5%tea polyphenol+0.2% nisin),C(0.2% nisin+0.3% lysozyme)and D(0.5% tea polyphenol+0.3% lysozyme)could prolong the preservation time of fresh T.flavidus to about 15 days.Fifteen samples from five groups of T.flavidus muscle under chilled storage were distributed in 187 genera,21 phyla,with dominant phyla including Proteobacteria,Actinobacteriota and Firmicutes.Pseudomonas,Achromobacter and Rhodococcus were the dominant genera.The compound biological preservatives could change the community structure of T.flavidus muscle,and Pseudomonas may be the specific spoil organisms of T.flavidus.This study provides theoretical references for the future research on preservation technology of T.flavidus.
