摘 要:研究探讨了脱胶脱脂对环肽提取产生的影响,以及不同测量方法之间测量蛋白含量的差异及环肽的抑菌效果。结果表明,温水脱胶时其脱胶量为8.43 g,1500 U/g的酶法脱胶时其脱胶量为9.01 g,100 U/g的酶法脱胶时其脱胶量为9.34 g。其次,1∶3和1∶6的料液比对蛋白提取量的影响不大,但是对提取的蛋白纯度的量影响较大,1∶6的料液比处理后蛋白纯度更高,为64.9%。凯氏定氮法得出在0.5 g的亚麻籽粉末样品中蛋白质含量约为0.23 g,考马斯亮蓝法得出环肽粗蛋白的提取率为64.9%,提取的蛋白纯度为58.32%。试验采用高效液相色谱法(HPLC)鉴定了提取的粗蛋白中含有氧化的环肽,并对提取出的环肽粗蛋白采用了柱层析法进行分离纯化,推测可能分离出了环肽。最后试验对提取的环肽粗蛋白进行了抑菌试验的检测,结果表明环肽粗蛋白对于大肠杆菌和金黄色葡萄球菌都具有一定的抑菌作用,且对前者的抑菌作用会更强。The effect of degluing and defatting on the extraction of cyclic peptide,the difference of protein content between different measurement methods and the bacteriostatic effect of cyclic peptide were studied.The results showed that the amount of degumming was 8.43 g in warm water,9.01 g in 1500 U/g enzymatic degumming and 9.34 g in 100 U/g enzymatic degumming.Secondly,the solid-liquid ratio of 1∶3 and 1∶6 had little influence on the protein extraction amount,but had a great influence on the protein purity.The protein purity of 1∶6 was higher than that of the treated protein,which was 64.9%.The protein content in 0.5 g flaxseed powder sample was about 0.23 g by Kjeldahl method.The extraction rate of cyclic peptide crude protein was 64.9%by Coomassie bright blue method,and the protein purity was 58.32%.High performance liquid chromatography(HPLC)was used to identify the oxidized cyclic peptide in the extracted crude protein,and column chromatography was used to separate and purify the extracted crude protein.It was speculated that the cyclic peptide might be isolated.In the end,the extracted cyclic peptide crude protein was tested by bacteriostatic experiment.The results showed that the cyclic peptide crude protein had certain bacteriostatic effect on Escherichia coli and Staphylococcus aureus,and the bacteriostatic effect on the former would be stronger.
