摘 要:为提高鼠李糖乳杆菌(Lactobacillus rhamnosus)R5胞外多糖(EPS)的产量,该研究以EPS产量为响应值,采用单因素试验及响应面试验对其产EPS的发酵条件进行优化,并采用Logistic、Luedeking-Piret、Luedeking方程建立鼠李糖乳杆菌产EPS发酵动力学模型。结果表明,鼠李糖乳杆菌R5产EPS的最优发酵条件为酵母粉添加量4%,葡萄糖添加量4%,接种量5%,发酵温度37℃,发酵时间24 h。在此优化条件下,鼠李糖乳杆菌R5的EPS产量最高,为1138.31 mg/L,是优化前的1.68倍。从构建的菌体生长、EPS产量及底物消耗的动力学模型可知,模型拟合曲线的相关系数均>0.98,拟合情况良好;预测值与试验值相近,相对误差均<15%,说明模型能很好地反映发酵过程规律。To increase the exopolysaccharide production by Lactobacillus rhamnosus R5,using the yield of exopolysaccharide(EPS)as the response value,the fermentation conditions of EPS production by L.rhamnosus R5 were optimized by single factor tests and response surface tests,and the fermentation kinetic model was established by Logistic,Luedeking-Piret,and Luedeking equations.The results showed that the optimum fermentation conditions for EPS production by L.rhamnosus R5 were yeast powder 4%,glucose 4%,inoculum 5%,fermentation temperature 37℃and time 24 h.Under the optimized condition,EPS production by L.rhamnosus R5 was the highest of 1138.60 mg/L,which was 1.68 times than before the optimization.The established kinetic models of cell growth,EPS production and substrate consumption indicated that the correlation coefficients of fitted curves of the models were all>0.98,and the fitting was good.The predicted values were closed to the experimental values,and the relative errors were all<15%,indicating that the model could reflect the fermentation process pattern well.
