摘  要:液体菌种已在黑木耳菌包工厂化生产中广泛应用并取得较好效果,但提高黑木耳液体菌种发酵生产效率仍是菌包生产企业的重要技术需求。为深入探究发酵营养条件、环境条件及其交互作用对黑木耳液体菌种生产效率的影响,为黑木耳液体菌种高效生产提供有益参考。试验以国家认定品种黑木耳2号为试验菌株,以发酵醪中菌丝量为响应值,采用经典的响应面法对液体菌种发酵过程进行研究。结果表明,发酵基质营养与环境条件对黑木耳菌丝生物量具有显著影响,试验范围均出现影响峰值;基质营养条件中的碳源浓度、环境条件中与溶氧水平密切相关的装液量和摇床转速都与其他发酵条件有很强的交互作用。结合单因素试验、响应面试验分析和验证试验,确定最佳配方为葡萄糖22.3 g/L、蛋白胨2.7 g/L、KH_(2)PO_(4)2.0 g/L、MgSO_(4)·7H_(2)O 1.5 g/L,同时添加羧甲基纤维素钠3 g/L,菌丝生物量达13.22 g/L,与预测值13.05 g/L相比,相对误差为1.3%;最佳发酵条件为温度30℃、时间7 d、摇床转速165 r/min、装液量105 mL(250 mL锥形瓶),菌丝生物量达到14.32 g/L,与预测值14.11 g/L相比,相对误差为1.49%。研究结果表明,此液体发酵培养基配方及调控条件有效可行、菌丝得率高。Liquid strains have been widely used in the factory production of Auricularia heimuer,and good effects have been achieved.However,improving the fermentation efficiency of Auricularia heimuer auricularia liquid strains is still an important technical requirement for the production enterprises of Auricularia heimuer.In order to further explore the effects of fermentation nutritional conditions,environmental conditions,and their interaction on the production efficiency of Auricularia heimuer liquid strain,and provide a useful reference for the efficient production of Auricularia heimuer liquid strain,in this paper,a national recognized variety Auricularia heimuer 2 was taken as the test strain,and the amount of mycelium in the fermentation mash as the response value,and the classical response surface method was adopted to conduct experimental research on the fermentation process of Auricularia heimuer liquid strain.The results showed that the nutrient and environmental conditions of fermentation substrate had significant effects on mycelium biomass of Auricularia heimuer,and the influence peak values were found in all experimental ranges.The carbon source concentration in the substrate nutrient condition,the liquid volume closely related to the dissolved oxygen level in the environment condition and the shaker speed all had strong interaction with other fermentation conditions.Combined with single factor test,response surface test analysis,and validation test,the optimal formula was determined as glucose 22.3 g/L,peptone 2.7 g/L,KH_(2)PO_(4)2.0 g/L,MgSO_(4)•7H_(2)O 1.5 g/L,and sodium carboxymethyl cellulose 3 g/L.The mycelial biomass reached 13.22 g/L,compared with the predicted value of 13.05 g/L,the relative error was 1.3%.The optimal fermentation conditions were as follows:temperature 30℃,time 7 d,shaker rotation speed 165 r/min,liquid volume 105 mL(250 mL conical flask),mycelial biomass was up to 14.32 g/L,compared with the predicted value of 14.11 g/L,the relative error was 1.49%.The results showed that the
