摘  要:该研究采用1,1-二苯基-2-三硝基苯肼(DPPH)自由基和羟自由基清除率试验测定混合植物果蔬酵素的体外抗氧化活性,并将Wistar大鼠随机分为正常组、模型组、中药组和酵素组,采用高脂饲料喂养联合腹腔注射链脲佐菌素(STZ)建立2型糖尿病(T2DM)大鼠模型,以体质量、饮水量、空腹血糖值(FBG)、口服葡萄糖耐量试验(OGTT)、血糖曲线下面积(AUC)及氧化应激指标为评价标准,研究混合植物果蔬酵素对T2DM大鼠血糖的影响。结果表明,发酵后,混合植物果蔬酵素的自由基清除能力提高,当含量为50 mL/L时,DPPH自由基清除率达到82.5%;当含量为200 mL/L时,羟自由基清除率达到89%。混合植物果蔬酵素可以控制T2DM大鼠体质量迅速下降,饮水量的增高,降低FBG、AUC,提高OGT,提高血清中超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、过氧化氢酶(CAT)水平,降低丙二醛(MDA)水平。综上,混合植物果蔬酵素具有抗氧化性,并可以改善T2DM大鼠血糖及氧化应激水平,具有预防或改善2型糖尿病的作用。The in vitro antioxidant activity of mixed fruit and vegetable Jiaosu was determined by 1,1-diphenyl-2-trinitrophenyl(DPPH)radical and hydroxyl radical scavenging assays.Wistar rats were randomly divided into normal group,model group,Chinese medicine group and Jiaosu group,and type 2 diabetes mellitus(T2DM)rat model was established by feeding high fat diet combined with peritoneal injection of streptozotocin(STZ).The effects of mixed fruit and vegetable Jiaosu on blood glucose in T2DM rats were investigated by using body mass,water intake,fasting blood glucose(FBG),oral glucose tolerance test(OGTT),area under curve of glucose(AUC)and oxidative stress indexes as evaluation criteria.The results showed that after the fermentation,the free radical scavenging ability of mixed fruit and vegetable Jiaosu was improved,and the scavenging rate of DPPH free radical reached 82.5%when the content was 50 ml/L,and the scavenging rate of hydroxyl radical reached 89%when the content was 200 ml/L.Mixed fruit and vegetable Jiaosu could control the rapid decrease of body mass,increase of water consumption in T2DM rats,reduce FBG and AUC,increase OGT,improve superoxide dismutase(SOD),glutathione(GSH)and catalase(CAT)levels in serum,and reduce MDA level.In conclusion,the mixed fruit and vegetable Jiaosu had antioxidant properties and could improve blood glucose and oxidative stress levels in T2DM rats,and had the effect of preventing or improving type 2 diabetes.
