摘 要:目的:比较国产和进口NBB培养基对啤酒有害菌的检测效果。方法:将干酪乳杆菌、短乳杆菌、四联球菌、戊糖片球菌、乳酸乳球菌乳脂亚种、植物乳杆菌植物亚种6株乳酸菌接种到NBB-B和NBB-A培养基中,28℃厌氧培养24~72 h,测定OD值,观察培养基变色、菌落生长状态等。同时,用不同浓度麦芽啤酒配制NBB培养基,确定配制培养基用啤酒的最适麦芽度。结果:6株乳酸菌在国产和进口NBB-B培养基中灵敏度和指示特征无明显差异。戊糖片球菌在国产NBB-A培养基培养48 h变色情况略差于进口NBB-A,培养72 h后,与进口NBB-A培养基无差异。通过不同麦芽度比对试验,采用10°P啤酒配制的NBB培养基菌株生长情况最好。结论:国产与进口NBB培养基在灵敏度、指示能力、菌落生长等方面无明显差异,国产NBB培养基可用于国内啤酒生产企业检测啤酒有害菌。Objective:To compare the detection effect of domestic and imported NBB culture media on harmful bacteria in beer.Method:Six strains of lactic acid bacteria,including Lactobacillus casei,Lactobacillus brevis,Micrococcustetragenus,Pediococcuspentosaceus,Lactococcus lactis subsp.cremoris,and Lactobacillus plantarum subsp.plantarum,were inoculated into NBB-B and NBB-A media.They were anaerobic cultured at 28℃for 24~72 h,and their OD values were measured.The discoloration of the media and the growth status of the colonies were observed.At the same time,prepare NBB culture medium with different concentrations of malt beer to determine the optimal malt content of the beer used for preparing the culture medium.Result:There was no significant difference in sensitivity and indicator characteristics between the 6 strains of lactic acid bacteria in domestic and imported NBB-B culture media.After 48 hours of cultivation on domestic NBB-A medium,the appearance of Pediococcuspentosaceus turned slightly worse than that on imported NBB-A.After 72 hours of cultivation,there was no difference compared to imported NBB-A medium.Through the comparison experiment of different malt degrees,the NBB culture medium prepared with 10°P beer showed the best growth performance.Conclusion:There is no significant difference in sensitivity,indicator ability,and colony growth between domestic and imported NBB culture media.Domestic NBB culture media can be used for detecting harmful beer bacteria in domestic beer production enterprises.
