摘 要:对分离自西藏灵菇的乳酸菌进行筛选,以获得对耐甲氧西林金黄色葡萄球菌(Methicillin-Resistant Staphylococcus aureus,MRSA)具有较强抑菌活性的菌株。以MRSA为指示菌对目的菌株进行筛选和鉴定,并对其安全性进行评价;测定该菌株抑菌活性物质对环境的耐受性。结果表明,菌株M3对MRSA的抑菌效果最好,抑菌圈直径可达18.47 mm;经鉴定该菌株为植物乳杆菌,其所产抑菌物质在–20~121℃内、30 min处理后仍具抑菌活性,对蛋白酶不敏感,对p H值敏感。乙酸乙酯可有效地萃取发酵液中的抑菌物质,经有机酸验证实验初步确定其成分为有机酸。植物乳杆菌M3不产生溶血现象,对氯霉素、头孢西丁、四环素和青链霉素敏感,不含可转移的质粒,具有潜在的安全性。该研究为新型抑菌剂的开发与利用提供了有效的理论基础。The lactic acid bacteria isolated from Tibetan kefir were screened to obtain strains with strong antimicrobial activity against(Methicillin-Resistant Staphylococcus aureus,MRSA).Taking MRSA as indicator bacteria,the target strain was screened and identified,and its safety was evaluated.The environmental tolerance of bacteriostatic substance from the strain was tested.The results showed that strain M3 had the best antibacterial effect on MRSA,with the diameter of antibacterial zone reaching 18.47 mm.The strain was identified as Lactiplantibacillus plantarum.The antibacterial substances produced by the strain still showed antibacterial activity after 30 min treatment at–20-121℃,and were insensitive to protease,but sensitive to pH.Ethyl acetate can effectively extract the bacteriostatic substances in fermentation broth,and they were preliminarily determined as organic acid by organic acid verification test.L.plantarum M3 did not produce hemolysis phenomenon,and was sensitive to chloramphenicol, cefoxitin, tetracycline and penicstreptomycin and did not contain transferable plasmids, which had potential safety. This study could provide a theoretical basis for the development and utilization of new bacteriostatic agents.
