摘 要:对梅卤多糖(PPM)进行了分离纯化,对梅卤多糖组分(PPM-Ⅱa)进行纯度分析及结构鉴定。利用DEAE-52纤维素阴离子柱层析法对PPM进行分离纯化得到4个组分,即PPM-Ⅰ、PPM-Ⅱ、PPM-Ⅲ和PPM-Ⅳ;经过凝胶渗透色谱筛选出PPM-Ⅱ组分为均一组分作为进一步纯化的对象;PPM-Ⅱ通过Sephadex G-100凝胶色谱柱纯化得到PPM-Ⅱa。PPM-Ⅱa主要是半乳糖、半乳糖醛酸和阿拉伯糖组成的酸性多糖,其含量分别为332832.01、25099.18、22801.19 mg/kg;经红外光谱与核磁共振氢谱判断,PPM-Ⅱa是含有β糖苷键的吡喃环酸性多糖,且刚果红试验结果表明PPM-Ⅱa存在三螺旋结构。The polysaccharides from plum bittern(PPM)was isolated and purified.The purity of plum bittern poly saccaride component(PPM-II a)was analyzed and its structure was identified.Four components,PPM-I,PPM-II,PPM-IIand PPM-IV,were isolated and purified by DEAE-52 cellulose anion column chromatography.After gel permeation chromatography,PPM-II was selected as a homogeneous component for further purification.PPM-II a was purified by Sephadex G-100 gel column.PPM-II a was mainly an acidic polysaccharide composed of galactose,galacturonic acid and arabinose,and its contents were 332832.01,25099.18 and 22801.19 mg/kg,respectively.According to the infrared spectrum and nuclear magnetic resonance hydrogen spectrum,PPM-II a was a pyranoic acid polysaccharide contai-ningβglycosidic bonds,and the Congo red test results showed that PPM-IIa had triple helix structure.
