摘  要:目的探究不同提取方法对羊栖菜多糖理化性质与体外生物活性的影响。方法以羊栖菜(Sargassum fusiforme)为研究对象,采用热水提取法(hot water extraction,HWE)、碳酸钠提取法(alkaline-assisted extraction,ALAE)、氯化钙提取法(calcium chloride-assisted extraction,CAAE)及微波辅助水提法(microwave-assisted water extraction,MAE)和超声辅助水提法(ultrasound-assisted water extraction,UAE)共5种方法提取羊栖菜多糖,分析不同提取方法对多糖的提取效果、理化性质、体外抗氧化和降血糖活性的影响。结果以水为溶剂进行提取,多糖得率相对较高,且微波和超声辅助可显著增强提取效果。ALAE提取的羊栖菜多糖分子量最低(924.96 k Da)且热稳定性最好(Tmax=255.84℃)。不同方法提取所得多糖的单糖组成类似,均主要由甘露糖、氨基葡萄糖、葡萄糖、阿拉伯糖、鼠李糖、半乳糖、半乳糖醛酸和岩藻糖所组成。此外,水提多糖表现出相对较强的体外抗氧化与降血糖活性。HWE提取的羊栖菜多糖具有最强的2,2’-联氨-双-3-乙基苯并噻唑啉-6-磺酸阳离子自由基[2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)ammonium salt cationic radical,ABTS+·]清除能力,其半数抑制浓度(half maximal inhibitory concentration,IC_(50))为1.33 mg/mL,UAE提取的多糖具有最强的1,1-二苯基-2-三硝基苯肼自由基(1,1-diphenyl-2-picrylhydrazyl radical,DPPH·)清除能力(IC_(50)=1.01 mg/mL),MAE提取的多糖表现出最强的还原力;且UAE提取所得多糖对α-葡萄糖苷酶抑制率高达(99.93±0.63)%。结论以水作为溶剂有利于羊栖菜多糖的提取,超声和微波辅助可有效强化提取效果,改善多糖生物活性。本研究为羊栖菜多糖的高效制备与开发应用提供了科学依据。Objective To study the effects of different extraction methods on the physicochemical properties and in vitro biological activities of Sargassum fusiforme polysaccharides.Methods A total of 5 extraction methods,including hot water extraction(HWE),alkaline-assisted extraction(ALAE),calcium chloride extraction(CAAE),microwave-assisted(MAE)and ultrasound-assisted(UAE)extraction,were used to extract polysaccharides from Sargassum fusiforme.The effects of different extraction methods on the polysaccharides yield,physicochemical properties,antioxidant and hypoglycemic activities in vitro were evaluated.Results The yield of polysaccharides was relatively high using water as solvent,and microwave and ultrasound could significantly enhance extraction efficiency.The polysaccharides extracted using ALAE had the lowest molecular weight(924.96 kDa)and the best thermal stability(Tmax=255.84℃).The monosaccharide composition of polysaccharides extracted by different methods was similar,and they were all mainly composed of mannose,glucosamine,glucose,arabinose,rhamnoose,galatose,galacturonic acid and fucose.In addition,the water-extracted polysaccharides showed excellent antioxidant and hypoglycemic activity in vitro.The polysaccharides extracted by HWE possessed the strongest 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)ammonium salt cationic radical(ABTS+ꞏ)scavenging ability,with an half maximal inhibitory concentration(IC_(50))of 1.33 mg/mL.The polysaccharides extracted by the UAE method exhibited outstanding 1,1-diphenyl-2-picrylhydrazyl radical(DPPHꞏ)scavenging ability(IC_(50)=1.01 mg/mL).The polysaccharides extracted by MAE had the best reducing power.Moreover,(99.93±0.63)%ofα-glucosidase could be inhibited by the polysaccharides extracted using MAE.Conclusion Water as solvent is beneficial to the extraction of polysaccharides from Sargassum fusiforme,and ultrasound and microwave can effectively enhance the extraction efficiency and improve the biological activities of polysaccharides.Thus,this study can
