摘 要:以鹿角菜为原料,采取乙醇溶剂提取法,以多酚提取率为指标,依据单因素试验结果,以BoxBehnken法优化鹿角菜多酚的提取工艺,并研究鹿角菜多酚的抗氧化活性。获得提取鹿角菜多酚的最优工艺条件为:乙醇浓度70%、料液比1∶12 g/mL、提取温度80℃、提取时间55 min,在此条件下多酚得率为5.21%。鹿角菜多酚对OH自由基的清除能力很高,当鹿角菜多酚浓度为16 mg/mL时达到最大值31.40%;对ABTS+自由基的清除能力较强,当鹿角菜多酚浓度为512 mg/mL时清除率达到最大值51.01%;对DPPH自由基的清除能力不高,当鹿角菜多酚浓度为64 mg/mL时最大值为34.13%;鹿角菜多酚的还原能力较弱但很稳定。The extraction process of polyphenols from Pelvetia siliquosa and its antioxidant activity were studied.Based on the single-factor test results,Box-Behnken method was used to optimize the process.The extraction rate of polyphenols was taken as the index.Response surface test results showed that the optimal extraction conditions were:ethanol concentration 70%,solid-liquid ratio 1∶12 g/mL,ultrasonic time 55 min,and ultrasonic temperature 80℃.Under these conditions,the yield of polyphenol was 5.21%.The antioxidant test showed that the scavenging ability of polyphenol from Pelvetia siliquosa was very high,reaching the maximum of 31.40% when the deer horn polyphenols concentration was 16 mg/mL,and the scavenging ability of ABTS+was stronger,reaching the maximum 51.01% when the deer horn polyphenols concentration was 512 mg/mL.The maximum of DPPH radical scavenging activity was 34.13% when the deer horn polyphenols concentration was 64 mg/mL,and the reduction ability of polyphenol was weaker but stable.
