摘 要:为有效降解烟梗提取物中的淀粉,利用脉冲安培-离子色谱法研究了α-淀粉酶、α-淀粉酶+糖化酶、α-淀粉酶+普鲁兰酶不同组合体系酶解梗膏中淀粉的效果,并对比了酶解后葡萄糖的生成量,分析了不同酶种类的效果差异。结果表明:单酶体系与双酶体系对于烟梗提取物梗膏中淀粉酶解效果、葡萄糖生成量的影响差异显著。α-淀粉酶+糖化酶、α-淀粉酶+普鲁兰酶双酶体系对淀粉的酶解效果和葡萄糖生成量均优于α-淀粉酶单酶体系。在酶用量方面,当淀粉降解率为50%时,α-淀粉酶+糖化酶体系需要α-淀粉酶365 U/g和糖化酶901 U/g,α-淀粉酶+普鲁兰酶体系需要α-淀粉酶365 U/g和普鲁兰酶815 U/g,而单酶体系需要α-淀粉酶7397 U/g,α-淀粉酶+普鲁兰酶双酶体系酶活单位用量最少;对应使用双酶体系酶解后葡萄糖质量百分数分别增加了6.18%和5.14%,而使用α-淀粉酶单酶体系酶解后,葡萄糖质量百分数仅增加4.71%,即α-淀粉酶+糖化酶体系葡萄糖的生成量最高。In order to effectively degrade starch in tobacco stem extract,the effect difference ofα-amylase,α-amylase+glucoamylase enzyme andα-amylase+pullulanase on starch degradation in tobacco stem extract was studied by ion chromatography with pulsed amperometric detection(PAD).Meanwhile,the contents of enzymolysis glucose were analyzed to compare the effects of different kind enzymes.The results showed that the enzymatic effects on starch and glucose production between a single-enzyme system and a dual-enzyme system were significantly different.The dual-enzyme system which consisted ofα-amylase with saccharifying enzyme andα-amylase with pullulanase had higher ratio of starch degradation and glucose production.In terms of enzyme dosage,when the starch degradation rate was 50%,α-amylase+saccharase system requiredα-amylase 365 U/g and saccharase 901 U/g,andα-amylase+Pullulanase system requiredα-amylase 365 U/g and Pullulanase 815 U/g,while single-enzyme system requiredα-amylase 7397 U/g.Theα-amylase+Pullulanase double enzyme system had the least amount of unit enzyme activity.The contents of glucose increased by 6.18%and 5.14%after enzymatic hydrolysis withα-amylase+saccharifying enzyme andα-amylase+pullulanase respectively,while the content of glucose increased by 4.71%after enzymatic hydrolysis with onlyα-amylase.The yield of glucose produced byα-amylase+saccharifying was the highest.
