摘 要:以玄米茶为原料,选取超声功率、超声时间、液料比、乙醇体积分数为自变量,利用单因素试验和响应面法优化玄米茶多酚的提取工艺。利用Fenton反应法、邻苯三酚自氧化法及DPPH比色法揭示玄米茶多酚的抗氧化活性。结果表明,超声辅助法提取玄米茶多酚的最佳工艺为超声功率350 W、超声时间80 min、料液比1︰32 g/mL、乙醇体积分数74%,在此条件下得到的提取率为34.76 mg/g。体外抗氧化试验表明,玄米茶多酚质量浓度0.5 mg/mL时,其对羟自由基(·OH)、超氧阴离子自由基(O_(2)^(-))和DPPH自由基的清除率分别为82.01%,82.57%和95.33%,表明玄米茶多酚具有良好的抗氧化活性。Polyphenols were extracted from Genmaicha by single factor test and response surface methodology.Ultrasonic power,ultrasonic time,liquid-material ratio and ethanol concentration were selected as experimental independent variables.The antioxidant activity of Genmaicha polyphenols was revealed by Fenton reaction,pyrogallol autoxidation and DPPH colorimetry.The results showed that the optimum condition of ultrasonic assisted extraction of Genmaicha polyphenols was as follows:ultrasonic power 350 W,ultrasonic time 80 min,solid-liquid ratio 1︰32 g/mL,and ethanol concentration 74%.Under this condition,the extraction rate was 34.76 mg/g.The antioxidant experiments in vitro showed that when the concentration of Genmaicha polyphenols was 0.5 mg/mL,the scavenging rates of hydroxyl radical(·OH),superoxide anion radical(O2-)and DPPH radical were 82.01%,82.57%and 95.33%,respectively,indicating that Genmaicha polyphenols had good antioxidant activity.
