摘  要:研究胶原三肽、普通胶原蛋白肽对光损伤(UVB)皮肤细胞的修复作用,以及不同胶原肽之间的修复差异。采用常规培养的人皮肤成纤维细胞,经UVB处理建模,以不同胶原肽干预UVB处理后的人皮肤成纤维细胞设计空白对照组,模型组,4种不同胶原肽的高、中、低剂量组,利用CCK8法检测细胞增殖的影响;利用凋亡试验检测细胞凋亡的影响;利用ELISA检测相关基因表达的影响:透明质酸、羟脯氨酸;利用荧光定量PCR检测细胞炎症相关基因表达:MMP1、MMP13、Ⅰ型前胶原(COL1a1)、Ⅲ型前胶原(COL3a1)和原弹性蛋白mRNA的表达。细胞增殖和细胞凋亡结果显示,胶原三肽及胶原蛋白肽对UVB处理后的人皮肤成纤维细胞均具有一定修复作用,且呈一定剂量相关性,其中效果显著的依次性为2^(#)胶原三肽>1^(#)胶原三肽>2^(#)胶原蛋白肽>1^(#)胶原蛋白肽。ELISA检测透明质酸和羟脯氨酸含量显示,胶原三肽、胶原蛋白肽均可以修复UVB损伤后人皮肤成纤维细胞生成细胞外基质的能力,且呈一定剂量相关性,其中效果显著的依次性为2^(#)胶原三肽>1^(#)胶原三肽>2^(#)胶原蛋白肽>1^(#)胶原蛋白肽。Western Blot检测蛋白表达发现,胶原三肽、胶原蛋白肽均有降低MMP-1、MMP-13,及提高COL1a1、COL1a2、原弹性蛋白信使RNA的趋势。胶原肽对光老化人皮肤成纤维细胞具有一定修复作用,其中胶原三肽的效果优于普通胶原蛋白肽。Investigate the effects of different collagen peptide and collagen tripeptide on photoaging in UVB-exposed human skin fibroblasts.Conventional cultured human skin fibroblasts were used for UVB treatment and modeling,and then different collagen peptide and collagen tripeptide were used to intervene human skin fibroblasts after UVB treatment.Blank control group,model group,high,medium and low dose groups of four different collagen peptides were designed.The effect of cell proliferation was detected by CCK8 method.Apoptosis assay was used to detect the effect of cell apoptosis.ELISA was used to detect the influence of related gene expression:hyaluronic acid and hydroxyproline;Fluorescence quantitative PCR was used to detect the expression of genes related to inflammation:MMP1,MMP13,type I pro-collagen(COL1a1),typeⅢprocollagen(COL3a1)and pro-elastin mRNA.The results of cell proliferation and apoptosis showed that collagen tripeptide and collagen peptide had repair effects on human skin fibroblasts treated with UVB in a dose-dependent manner,and the significant effect was 2^(#)collagen tripeptide1^(#)collagen tripeptide2^(#)collagen peptide1^(#)collagen peptide.ELISA analysis of hyaluronic acid and hydroxyproline showed that both collagen tripeptide and collagen peptide could repair the ability of skin fibroblasts to generate extracellular matrix after UVB injury,and in a certain dose-dependent manner,the significant effect was 2^(#)collagen tripeptide1^(#)collagen tripeptide2^(#)collagen peptide1^(#)collagen peptide.Western Blot analysis showed that collagen tripeptide and collagen peptide both decreased MMP-1 and MMP-13,and increased COL1a1,COL1a2 and pro-elastin mRNA.Collagen peptide and collagen tripeptide had a certain repair effect on skin fibroblasts of photoaged people,and the effect of collagen tripeptide was better than that of ordinary collagen peptide.
