摘 要:为提高本土优良酿酒酵母的生物量并降低培养基的使用成本,以优选自宁夏贺兰山东麓葡萄酒产区的本土酿酒酵母CEC D3-6为研究对象,分别以甘蔗糖蜜和玉米浆作为培养基的碳源和氮源,采用单因素试验和响应面相结合的方法,优化了供试菌株的半合成发酵培养基。结果表明,甘蔗糖蜜、玉米浆和七水合硫酸镁是影响酿酒酵母生物量的主效因素(置信度大于95%),当培养基中的甘蔗糖蜜、玉米浆和七水合硫酸镁含量分别为102.49、30.30、6.12 g·L^(-1)时,菌体干质量达到8.37 g·L^(-1),比优化前的培养基提高了2.99倍。研究结果为本土优选酿酒酵母的高密度培养,及本土酿酒酵母高活性酵母干粉生产成本的降低奠定了基础。In order to improve the biomass of excellent local Saccharomyces cerevisiae and reduce the cost of culture medium,Saccharomyces cerevisiae CEC D3-6 from the wine producing area in the eastern foot of Helan Mountain as research object,using sugarcane molasses and corn steep liquor as the carbon and nitrogen sources of culture medium,and the method of combining single factor test and response surface was adopted,optimized the semi synthetic fermentation medium of Saccharomyces cerevisiae.The results showed that sugarcane molasses,corn steep liquor,and magnesium sulfate heptahydrate were the main factors affecting the biomass of Saccharomyces cerevisiae(with a confidence level greater than 95%).When the contents of sucrose molasses,corn steep liquor,and magnesium sulfate heptahydrate inculture medium were 102.49,30.30,6.12 g·L^(-1),respectively,the dry weight reached 8.37 g·L^(-1),2.99 times higher than the optimized medium.The results of this study lay a foundation for the high-density cultivation of native selected Saccharomyces cerevisiae and reducing the production cost of native selected Saccharomyces cerevisiae highly active yeast dry powder.
