摘 要:该研究以白酒丢糟为研究对象,采用单因素试验及正交试验对复合酶制备微晶纤维素(MCC)的工艺条件进行优化,并对MCC的结构进行表征。以乳酸乳球菌(Lactococcus lactis)生物量为评价指标,以MCC制备过程中产生的废水替代乳酸链球菌产Nisin培养基中的蒸馏水,并采用单因素试验及响应面法对其培养基组分进行优化。结果表明,最佳MCC制备工艺参数为:纤维素酶添加量24 U/g,酶解时间9 h,料液比1∶20(g∶m L)。在此条件下,MCC的聚合度(DP)为118,其结构呈棒状,粒径约为20μm;最佳乳酸乳球菌产Nisin培养基组分为:在1 000 m L废水添加酵母粉8 g、蛋白胨8 g、蔗糖9 g、KH2PO419 g、Mg SO40.15 g,在此优化条件下,菌株生物量为6.3×107CFU/m L,Nisin效价为732.52 IU/m L。In this study,using Baijiu distiller's grains as research object,the preparation technology of microcrystalline cellulose(MCC) and Nisin by the multi-enzyme was optimized by single factor tests and orthogonal tests,and the structure of MCC was characterized.Using the biomass of L.lactis as evaluation index,the distilled water in Nisin culture medium was replaced by the wastewater produced from the MCC preparation process,and the components of the medium were optimized by single factor tests and response surface method.The results showed that the optimal preparation process parameters of MCC were as follows:cellulase addition 24 U/g,enzymolysis time 9 h,solid-liquid ratio 1∶20(g∶ml).Under these conditions,the degree of polymerization(DP) of MCC was 118,with rod-like structure,and the particle size was about 20 μm.The optimal medium components for Nisin production by L.lactis were wastewater 1 000 ml,yeast powder 8 g,peptone 8 g,sucrose 9 g,KH2PO419 g and Mg SO40.15 g.Under the optimized conditions,the biomass of strain was 6.3×107CFU/ml,and the titer of Nisin was 732.52 IU/ml.
