摘  要:探究化学修饰对木聚糖酶(XY)相对活性的影响,以XY相对活性为指标,利用丁二酸酐、乙酸酐、马来酸酐(MA)和邻苯二甲酸酐对XY进行化学修饰。采用单因素试验筛选出最优酸酐修饰剂,并进行响应面法优化酸酐修饰条件。结果表明:MA为最优修饰酸酐,最佳修饰条件为修饰温度14℃、修饰时间4h、pH7.0、MA添加量40%(以XY质量计)。在此条件下,马来酸酐修饰木聚糖酶(MA-XY)的相对活性为154.62%±1.24%。To explore the effect of chemical modification on xylanase(XY)relative activity,XY was chemical modified with succinic anhydride,acetic anhydride,maleic anhydride(MA)and phthalic an-hydride based on XY relative activity.The optimal anhydride modifier was selected by single factor test,and the modification conditions were optimized by response surface methodology.The results showed that MA was the optimal modification anhydride,and the optimal modification conditions were as follows:modification temperature 14℃,modificationtime4h,pH 7.0,and MA addition40%(basedonXY mass).Under these conditions,the relative activity of maleic anhydride modified xylanase(MA-XY)was 154.62%±1.24%.
