摘 要:对臭常山内生真菌Diaporthe orixae sp.nov.挥发性成分及其生物活性进行分析,以臭常山内生真菌Diaporthe orixae sp.nov.为研究对象,采用顶空固相微萃取结合气相色谱质谱联用(HS-SPME-GC-MS)的方法分析其挥发性成分,并进行生物活性测试。从Diaporthe orixae sp.nov.发酵液、菌丝体、臭常山茎对照药材中共鉴定出203种化合物,它们主要为醇类、酯类、烃类、酸类、酮类、酚类、醛类和其他类,Diaporthe orixae sp.nov.发酵液、菌丝体、臭常山茎对照药材的成分种类和相对含量存在差异,其中醇类、酯类、烃类化合物总占比较大,共有的挥发性成分有12种。抗氧化活性显示菌液粗提物对DPPH自由基、羟基自由基的IC_(50)值分别为0.05152和0.0479 mg/mL,其清除能力均大于菌体粗提物;对α-葡萄糖苷酶的抑制作用表明,Diaporthe orixae sp.nov.具有潜在的降血糖活性。上述结果为臭常山内生真菌的开发利用提供新思路,为真菌来源的活性天然产物的开发应用提供基础。To analyze the volatile components of endophytic fungus from Orixa japonica and investigate its bioactivity,using Diaporthe orixae sp.nov.from Orixa japonica as raw material,headspace solid phase microextraction combined with gas chromatography-mass spectrometry(HS-SPME-GC-MS)was used to analyze its volatile components and their biological activities were tested.A total of 203 compounds were identified from Diaporthe orixae sp.nov.fermentation broth,mycelium and rhizome of Orixa japonica Thunb.,which were mainly alcohols,esters,hydrocarbons,acids,ketones,phenols,aldehydes and other classes,there were differences in the composition types and relative contents in Diaporthe orixae sp.nov.fermentation broth,mycelium and rhizome of Orixa japonica Thunb.,among which alcohols,esters and hydrocarbons accounted for a relatively large proportion,and there were 12 volatile components in common.The IC_(50) values of DPPH free radical and hydroxyl free radical of the crude extract of Diaporthe orixae sp.nov.fermentation broth were 0.05152 and 0.0479 mg/mL,respectively.The scavenging ability of the crude extract of Diaporthe orixae sp.nov.fermentation broth was higher than that of the crude extract of bacterial body;the inhibition ofα-glucosidase showed that Diaporthe orixae sp.nov.had potential hypoglycemic activity.The above results provide a new idea for the development and utilization of endophytic fungi from Orixa japonica Thunb.,and provide a basis for the development and application of active natural products derived from fungi.
