摘  要:采用膜滤方法从紫薯淀粉加工副产物中制备花色苷,采用单因素试验探讨柠檬酸浓度、提取时间、料液比对提取效果的影响,确定紫薯花色苷较适宜的提取条件,即柠檬酸浓度2%,提取时间60 min,料液比1:6(g/mL)。通过小鼠饲喂试验,测定血清中总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)和高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)等指标以探究紫薯花色苷(purple potato anthocyanins,PSPAs)对高脂膳食小鼠血脂的影响。结果表明,PSPAs能够显著降低高脂膳食小鼠血清中TC、TG、LDL-C含量及升高HDL-C含量,且呈现一定的量效关系。与模型对照组相比,低剂量组(13 mg/kg)的综合降血脂效果最好,能使小鼠血清TC、TG、LDL-C含量分别下降15.10%,21.77%和33.33%,HDL-C升高6.29%,且低剂量的PSPAs能使高脂膳食小鼠血清TG、HDL-C含量恢复到正常水平,效果与阳性对照组无差异。Anthocyanins were prepared from purple sweet potato starch processing by-products by membrane filtration method.The effects of citric acid concentration,extraction time and solid-liquid ratio on the extraction effect were investigated by single factor experiment.The suitable extraction condition of purple sweet potato anthocyanins was determined as citric acid concentration of 2%,extraction time of 60 min,and solid-liquid ratio of 1︰6(g/mL).Through the mouse feeding experiment,total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C)and high density lipoprotein cholesterol(HDL-C)in serum were measured to investigate the effects of purple potato anthocyanins(PSPAs)on blood lipids in mice fed a high-fat diet.The results showed that PSPAs could significantly reduce the contents of TC,TG and LDL-C,and increase the content of HDL-C in the serum of high-fat diet mice,and showed a certain dose-effect relationship.Compared with the model control group,the comprehensive lipid-lowering effect of the low-dose group(13 mg/kg)was the best,which could reduce the serum TC,TG and LDL-C content of mice by 15.10%,21.77%and 33.33%,and increase HDL-C by 6.29%,respectively.The low-dose PSPAs could restore the serum TG and HDL-C content of high-fat diet mice to normal levels,and the effect was not different from that of the positive control group.
