摘 要:目的探究小麦肽、大豆肽和海参肽对H_(2)O_(2)诱导的L929细胞氧化应激损伤的保护作用及促细胞增殖能力比较。方法首先通过测定L929细胞的增殖率方法来确定L929细胞的浓度及H_(2)O_(2)的浓度,建立L929细胞H_(2)O_(2)损伤模型,测定3种肽对L929细胞的氧化损伤保护能力,再通过测定细胞周期及细胞凋亡率的方法来比较3种肽的促细胞增殖能力。结果海参肽的氧化损伤保护能力优于大豆肽和小麦肽;200μg/mL大豆肽和海参肽处理后的成纤维细胞增殖指数相比于空白对照都有显著增加,即大豆肽和海参肽可以通过影响细胞周期提高增殖指数而促成纤维细胞增殖,且同浓度海参肽的促成纤维细胞增殖能力优于同浓度的大豆肽,而小麦肽对细胞周期的影响无显著性差异。海参肽的抑制成纤维细胞凋亡作用最强,经200μg/mL海参肽作用后,细胞的凋亡率由空白对照的的(3.59±0.11)%降至(1.89±0.09)%。结论海参肽对细胞氧化损伤保护及促细胞增殖能力最强,大豆肽次之。Objective To investigate the protective effects of wheat peptide,soybean peptide and sea cucumber peptide on H_(2)O_(2)-induced oxidative stress injury in L929 cells and the comparison of cell proliferation promoting ability.Methods Firstly,the concentration of L929 cells and the concentration of H_(2)O_(2)were determined by measuring the proliferation rate of L929 cells,and the H_(2)O_(2)damage model of L929 cells was established,and the abilities of 3 kinds of peptides to protect L929 cells from oxidative damage were measured.Then,the cell cycle and apoptosis rate were measured to compare the cell proliferation promoting abilities of 3 kinds of peptides.Results The oxidative damage protection ability of sea cucumber peptides was stronger than that of soybean peptides and wheat peptides;the proliferation indices of fibroblasts treated with 200μg/mL soybean peptide and sea cucumber peptide had significant increase compared with that of the blank control,i.e.,soybean and sea cucumber peptides could improve proliferation indices by that is,soybean peptide and sea cucumber peptide could contribute to fibroblast proliferation by influencing the cell cycle and increasing the proliferation index,and the ability of the same concentration of sea cucumber peptide to contribute to fibroblast proliferation was better than that of the same concentration of soybean peptide,while the effect of wheat peptide on the cell cycle was not significantly different.Sea cucumber peptide had the strongest inhibitory effect on apoptosis of fibroblasts,and the apoptosis rate of cells decreased from(3.59±0.11)%in the blank control to(1.89±0.09)%after 200μg/mL sea cucumber peptide.Conclusion Sea cucumber peptide has the strongest protective effect against oxidative damage and cell proliferation-promoting ability,followed by soybean peptide.
