摘 要:为了评价不同桑叶提取物对α-淀粉酶、α-葡萄糖苷酶和蔗糖酶的抑制作用,采用不同工艺制备得到不同桑叶提取物并测定其成分含量,分别建立α-淀粉酶、α-葡萄糖苷酶和蔗糖酶的抑制测定方法,并测定不同桑叶提取物的抑制率和半抑制浓度(semi-inhibitory concentration,IC_(50))。结果显示不同桑叶提取物对α-淀粉酶有一定的促进作用,桑叶1-脱氧野尻霉素(1-deoxynojirimycin,DNJ)提取物促进作用最小。对α-葡萄糖苷酶有明显的抑制作用,桑叶DNJ提取物抑制作用最强。对蔗糖酶有不同程度的抑制作用,桑叶黄酮提取物、桑叶DNJ粗提取物抑制作用最强。表明不同桑叶提取物其活性成分标示含量不同,对α-淀粉酶、α-葡萄糖苷酶和蔗糖酶的抑制作用不同,对桑叶提取物辅助降血糖产品开发应用具有指导意义。In order to evaluate the inhibitory effects of different mulberry leaf extracts onα-amylase,α-glucosidase and sucrase,different mulberry leaf extracts were prepared by different extracting processes,and their components were determined.The inhibitory detection methods ofα-amylase,α-glucosidase and sucrase were established,and the inhibitory rate and semi-inhibitory concentration(IC_(50))of the extracts of mulberry leaves were analyzed.The results showed that all mulberry leaf extracts had certain stimulus effects onα-amylase,with DNJ(1-deoxynojirimycin)extract showing the weakest stimulus effect.They demonstrated significant inhibitory effects onα-glucosidase,DNJ extract showed the strongest inhibitory effect onα-glucosidase.They exhibited varying degrees of inhibition on sucrase,with mulberry leaf flavonoids extract and mulberry leaf DNJ crude extract showing the strongest inhibitory effects on sucrase.The study showed that different mulberry leaf extracts had different levels of active ingredient contents,leading to different inhibitory effects onα-amylase,α-glucosidase,and sucrase.These findings help for the development and application of mulberry leaf extract-based hypoglycemic products.
