大豆蛋白和大豆肽对小鼠焦虑样行为的作用研究

摘  要:为研究大豆蛋白和大豆肽对慢性不可预测轻度应激(chronic unpredictable mild stress,CUMS)诱导的小鼠焦虑样行为的影响,选用40只6~7周龄雄性C57BL/6J小鼠,随机分为空白组、模型组、大豆蛋白组和大豆肽组。通过CUMS诱导小鼠产生焦虑样行为,并在第53天进行高架十字迷宫实验,第54天收集脑组织样本检测小胶质细胞的激活情况、炎症因子和脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)的水平,以及BDN F-Trk B-CREB通路上相关蛋白的表达。结果表明:与空白组相比,CUMS可使小鼠在开臂中的时间和进入开臂的次数占比显著降低,Ib a-1阳性细胞数量显著增加,I L-1β水平显著上升,BDNF、p-TrkB、p-CREB表达显著下降(P<0.05);与模型组相比,大豆蛋白组中小鼠进入开臂的次数占比为模型组的1.4倍,同时I L-6、I L-1β、TN F-α和IF N-γ水平分别降低了21.5%、34.2%、10.9%和32.5%,BDNF蛋白表达提高了39.8%(P<0.05)。大豆肽组中小鼠在开臂中的时间和进入开臂的次数分别为模型组的2.0倍和1.4倍,同时小鼠海马区、CA1和CA3区Ib a-1阳性细胞数量分别下降了39.0%、43.2%和40.8%,I L-10水平提高了23.3%,I L-1β和TN F-α水平分别降低了26.8%和13.7%,BDNF水平提高了22.0%,BDNF和p-TrkB蛋白表达分别提高了63.0%和42.5%(P<0.05)。研究结果表明,大豆蛋白和大豆肽营养干预均可缓解神经炎症、增强神经营养功能,从而发挥抗焦虑活性,其中大豆肽效果更佳。研究旨在为抗焦虑食品的研发提供理论参考。To study the effects of soybean protein and soybean peptides on anxiety-like behavior induced by chronic unpredictable mild stress(CUMS)in mice,forty male C57BL/6J mice aged 6-7 weeks were selected and randomly divided into a control group,a model group,a soybean protein group,and a soybean peptides group.The mice were induced to develop anxiety-like behaviors by CUMS and were subjected to an elevated plus maze test on 53 d.Brain tissue samples were collected on 54 d to examine the activation of microglia,the levels of inflammatory factors and brain-derived neurotrophic factor(BDNF),and the expression of related proteins on the BDN F-Trk B-CREB pathway.The results showed that compared with the control group,CUMS resulted in a significant decrease in the percentage of time spent in the open arms and numbers of entries into the open arms,a significant increase in the number of Ib a-1 positive cells,a significant increase in the level of the I L-1β,and a significant decrease in the expression of BDNF,p-TrkB,and p-CREB in mice(P<0.05).Compared with the model group,the percentage of entries into the open arms of mice in the soybean protein group was 1.4 times higher than that in the model group,while I L-6,I L-1β,TN F-α,and IF N-γlevels were reduced by 21.5%,34.2%,10.9%,and 32.5%,respectively,and BDNF protein expression was increased by 39.8%(P<0.05).In the soybean peptides group,the percentage of time spent in the open arms and numbers of entries into the open arms of mice were 2.0 and 1.4 times higher than those in the model group,while the number of Ib a-1-positive cells in the hippocampus,CA1,and CA3 regions of the mice decreased by 39.0%,43.2%,and 40.8%,respectively,the levels of I L-10 were increased by 23.3%,and the levels of I L-1βand TN F-αwere decreased by 26.8%and 13.7%,respectively.BDNF levels increased by 22.0%,and BDNF and p-TrkB protein expression increased by 63.0%and 42.5%,respectively(P<0.05).The results of the study suggested that both soybean protein and soybean peptides nutritional intervent
