摘 要:建立基于柱前衍生化的超高效液相色谱-质谱方法检测不同产地黑蒜中硒代蛋氨酸含量。建立的检测方法对硒代蛋氨酸检出限为4.844 ng/mL,加样回收率在71.4%~91.4%之间,满足分析检测要求。6个产地黑蒜中硒代蛋氨酸含量差异明显,其中青海和江苏产含量最高。使用正交偏最小二乘法对6个不同产地的黑蒜核磁共振氢谱数据进行鉴别分析,不同产地黑蒜具有明显的聚类关系。差异代谢物主要包括氨基酸、植物固醇和糖类物质。黑蒜硒代蛋氨酸与核磁共振氢谱中的缬氨酸信号峰具有较高的相关性(r=0.78)。建立的异硫氰酸苯酯柱前衍生化超高效液相色谱-质谱检测硒代蛋氨酸方法稳定可靠。核磁共振氢谱技术可鉴别分析不同产地黑蒜,且缬氨酸峰的强度与硒代蛋氨酸含量相关性较高。There is a determination of selenomethionine in black garlic from different habitats by ultra-high performance chromatography-mass spectrometry based on pre-column derivatization.The results showed that the detection limit of selenomethionine is 4.844 ng/mL,and the recovery rate was 71.4%-91.4%.There are obvious differences in selenomethionine content in black garlic from six producing areas,among which the content in Qinghai and Jiangsu is different.Using orthogonal partial least squares method to identify black garlic from 6 different habitats,the black garlic from different habitats has obvious clustering relationship.Differential metabolites mainly include amino acids,phytosterols and sugars.Selenomethionine in black garlic has a high correlation with valine peak in 1HNMR spectrum(r=0.78).The established method is stable and reliable for the determination of selenomethionine in black garlic by pre-column derivatization of phenyl isothiocyanate and ultra-performance liquid chromatography-mass spectrometry.It can be applied to the determination of selenomethionine in black garlic from different habitats.The high correlation between the intensity of valine peak and the content of selenomethionine is helpful to calculate the relative content of selenomethionine in black garlic.
