摘  要:利用壳聚糖对羊栖菜多糖进行脱蛋白处理。以多糖保留率和脱蛋白率为指标,在单因素试验的基础上,通过响应面法优化羊栖菜多糖的脱蛋白工艺,并评价脱蛋白后羊栖菜多糖的抗氧化活性。结果表明:羊栖菜多糖的最佳脱蛋白工艺条件为羊栖菜多糖溶液与壳聚糖溶液体积比1∶2.00、壳聚糖分子质量500 kDa、温度5℃、pH 7.0。在此条件下,多糖保留率为78.23%,脱蛋白率为73.14%。体外抗氧化试验结果显示,脱蛋白后的羊栖菜多糖对DPPH自由基和羟基自由基的最大清除率可分别达到70.27%和70.19%,表明脱蛋白后的羊栖菜多糖均具有一定的抗氧化活性。Sargassum fusiforme polysaccharide was deproteinized by chitosan.Taking polysaccharide re-tention rate and deproteinization rate as indexes,the deproteinization process of Sargassum fusiforme poly-saccharide was optimized by response surface methodology based on single factor experiments,and the antioxidant activities of Sargassum fusiforme polysaccharide after deproteinization were evaluated.The re-sults showed that the optimal deproteinization process conditions for Sargassum fusiforme polysaccharide were volume ratio of Sargassum fusiforme polysaccharide solution to chitosan solution 1:2.00,molecular mass of chitosan 500 kDa,temperature 5℃,and pH 7.0.Undertheseconditions,polysaccharidereten-tion rate was 78.23%,and deproteinization rate was 73.14%.The results of in vitro antioxidant test showed that the maximum scavenging rate of DPPH radicals and hydroxyl radicals of the Sargassum fusi-forme polysaccharide after deproteinization could reach 70.27%and 70.19%,respectively,indicating that Sargassum fusiforme polysaccharide after deproteinization possessed moderate antioxidant activities.
