摘 要:目的:为核桃加工废弃物分心木的综合开发利用提供理论依据,减少资源浪费。方法:以多酚得率为评价指标,考察复合酶比例、时间、酶浓度、温度、pH值、料液比对分心木多酚提取得率的影响,并评价其抗氧化、抑制亚硝化和降糖能力。结果:在复合酶质量比(果胶酶:纤维素酶)2:1、提取时间3 h、复合酶浓度1.5%、提取温度70℃、pH6、料液比1:70的提取条件下,分心木多酚得率最高为193.21 mg/g。分心木多酚对1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基、2,2’-联氮-双(3-乙基苯并噻唑啉-6-磺酸)(2,2’-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS)阳离子自由基和铁离子还原力的半数抑制浓度(IC_(50))值分别为13.27、20.99μg/mL和0.22 mg/mL,对亚硝酸钠清除和α-葡萄糖苷酶抑制的IC_(50)值分别为0.38、0.29 mg/mL,且分心木多酚的生物活性与其浓度呈剂量依赖效应。结论:复合酶法能有效提取核桃分心木中的多酚,分心木多酚具有良好的抗氧化、亚硝酸盐清除活性和降糖能力。Objective:To provide a theoretical basis for the comprehensive development and utilization of walnut processing wastes,and to reduce the waste of resources.Methods:Taking the polyphenol yield as the evaluation index,the effects of composite enzyme ratio,time,enzyme concentration,temperature,pH,and material-liquid ratio on the polyphenol extraction rate of Diaphragma juglandis Fructus were investigated,and its antioxidant,nitrosation inhibition,and glucose-lowering ability were evaluated.Results:The highest polyphenol yield of 193.21 mg/g was obtained from Diaphragma juglandis Fructus under the extraction conditions of complex enzyme mass ratio(pectinase:cellulase)of 2:1,extraction time of 3 h,complex enzyme concentration of 1.5%,extraction temperature of 70℃,pH 6,and material-liquid ratio of 1:70.The half scavenging concentration(IC_(50))values for 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical,2,2’-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS)radical cation and iron reducing power were 13.27,20.99μg/mL and 0.22 mg/mL,respectively.IC_(50)for nitrite scavenging andα-glucosidase were 0.38 mg/mL and 0.29 mg/mL,respectively.The biological activity of Diaphragma juglandis Fructus polyphenols showed a dose-dependent effect with its concentration.Conclusion:The composite enzyme method can effectively extract polyphenols from Diaphragma juglandis Fructus,and the polyphenols have good antioxidant,nitrite scavenging activity and hypoglycemic ability.
