摘  要:建立基于稳定同位素比值质谱技术测定红薯淀粉和木薯淀粉δ^(15)N的方法。选取我国红薯、木薯主产区的高淀粉含量的红薯和木薯品种为主要研究对象,采用湿法水洗工艺制备红薯和木薯淀粉试样,通过提取其中的粗蛋白质将微量的氮富集。通过δ^(15)N在仪器上的信号值来优化提取条件,最佳提取工艺条件为超声提取时间2 h、料液比3∶45(g/mL)、用乙醇2次洗涤沉淀。利用该方法对红薯和木薯样品的粗蛋白质进行δ^(15)N的测定,结果发现,红薯淀粉的δ^(15)N在20.68%~55.05%之间,木薯淀粉δ^(15)N在63.77%~117.64%之间,两者差异明显。该研究为红薯和木薯淀粉的鉴别和产地溯源的研究提供新方法。A method for the determination ofδ^(15)N in sweet potato starch and cassava starch based on stable elemental analyzer isotope ratio mass spectrometry(EA-IRMS)was established.Sweet potato and cassava varieties with high starch content in the main producing areas of sweet potato and cassava in China were selected as the main research objects.Sweet potato starch and cassava starch samples were prepared by wet washing process,and then trace nitrogen was enriched by extracting crude protein to achieve the purpose of enhancingδ^(15)N signal.Through the signal feedback ofδ^(15)N on EA-IRMS,the results showed that the optimum extraction condition was as follows:ultrasonic extraction time was 2 h,solid-liquid ratio was 3∶45(g/mL),and rinse the proteinprecipitate with ethanol twice.The results showed that theδ^(15)N of crude protein in sweet potato starch and cassava starch extracted by this method were significantly different.Theδ^(15)N value of sweet potato starch was 20.68%-55.05%,and theδ^(15)N value of cassava starch was 63.77%-117.64%.It provided a new method for the identification and origin traceability of sweet potato and cassava starch.
