摘 要:以虎奶菇为原料,采用超声波辅助乙醇溶液提取虎奶菇多酚。以多酚得率为指标,在单因素试验基础上,通过响应面法优化提取工艺,并对提取物的抗氧化活性进行测定。结果表明:最佳提取工艺为超声功率300 W、提取时间56 min、乙醇体积分数44%、提取温度59℃,在此工艺条件下,多酚得率为(1.022±0.013)mg/g。抗氧化试验测定发现,虎奶菇多酚具有较好的抗氧化活性,其清除DPPH自由基和羟基自由基的IC50分别为2.010 mg/mL和0.430 mg/mL。Using Pleurotus tuber-regium(Fr.)Sing as raw material,polyphenols were extracted by ultrasound-assisted ethanol solution.Taking the yield of polyphenols as the index,the extraction process was optimized by response surface methodology based on single factor experiments,and the antioxidant activity of the extract was determined.The results showed that the optimal extraction process were ultrasound power 300 W,extraction time 56 min,ethanol volume fraction 44%,and extraction temperature 59℃.Under these process conditions,the polyphenol yield was(1.022±0.013)mg/g.The antioxidant test showed that polyphenols from Pleurotus tuber-regium(Fr.)Sing had good antioxidant activity,and the IC_(50)of the scavenging DPPH free radicals and hydroxyl free radicals were 2.010 mg/mL and 0.430 mg/mL,respectively.
