摘 要:为优化超声波辅助双水相提取鸡腿堇菜粗多糖工艺并初步研究其抗氧化活性,以鸡腿堇菜为试材,鸡腿堇菜粗多糖得率为响应值,考察双水相PEG6000/K_(2)HPO_(4)质量比、超声提取温度、超声提取时间及料液比对鸡腿堇菜粗多糖得率的影响,采用响应面试验优化鸡腿堇菜粗多糖的超声辅助双水相提取工艺,并通过测定自由基清除能力考察鸡腿堇菜粗多糖的抗氧化活性。结果表明:鸡腿堇菜粗多糖提取的最优工艺为:以PEG6000/K_(2)HPO_(4)(2.0 g:1.6 g→10 mL)双水相体系为提取溶剂,料液比(m_(鸡腿堇菜粗粉):V_(双水相体系))为1:29(g/mL),摇匀浸润30 min后,超声辅助提取,超声提取温度58℃,超声提取时间80 min,鸡腿堇菜粗多糖得率为6.79%±0.11%;抗氧化试验结果表明,鸡腿堇菜粗多糖对1,1-二苯基-2-三硝基苯肼(DPPH)自由基及羟基自由基清除能力随着质量浓度的增大而增强,当鸡腿堇菜粗多糖质量浓度为6 mg/mL时,对DPPH自由基清除率为70.13%,当鸡腿堇菜粗多糖质量浓度为10 mg/mL时,对羟基自由基清除率可达69.21%,表明鸡腿堇菜粗多糖具有一定的体外抗氧化活性。研究结果为鸡腿堇菜植物资源的开发与利用提供了理论依据及技术参考。This study aimed to optimize the ultrasonic-assisted aqueous two-phase extraction technology of crude polysaccharides from Viola acuminata Ledeb.and investigate the free radicals scavenging activity.The Viola acuminata Ledeb.was used as the experimental material,the effects of aqueous two-phase PEG6000/K_(2)HPO_(4) mass ratios,ultrasonic extraction temperatures,ultrasonic extraction time,and solid-liquid ratios on the yields of crude polysaccharides were investigated.Besides,response surface methodology was used to optimize the ultrasonic-assisted aqueous two-phase extraction technique of the crude polysaccharides,and the antioxidant activity was determined by free radicals scavenging ability experiments.The results showed that the optimum extraction conditions for crude polysaccharides from Viola acuminata Ledeb.were determined as follows:the extraction solvent of PEG6000/K_(2)HPO_(4)(2.0 g∶1.6 g→10 mL)aqueous twophase system,solid-liquid ratio(Viola acuminata Ledeb.powder∶aqueous two-phase system)of 1∶29(g/mL),mixing and infiltration time of 30 min,ultrasonic-assisted extraction temperature of 58℃,and extraction time of 80 min.Under these conditions,the yield of crude polysaccharides from Viola acuminata Ledeb.was 6.79%±0.11%.The antioxidant experiment results showed tha the scavenging effects of the crude polysaccharides on 1,1-diphenyl-2-picrylhydrazyl(DPPH)and hydroxyl free radicals were ascended with the increase of mass concentrations.The scavenging rate of DPPH free radical was 70.13%with the crude polysaccharides mass concentration of 6 mg/mL,and the scavenging rate of hydroxyl radical was 69.21%with the mass concentration of 10 mg/mL,which indicated that the crude polysaccharides had certain antioxidant activities in vitro.The results could provide theoretical basis and technical references for the development and utilization of Viola acuminata Ledeb.
