摘 要:为揭示产己酸功能菌液在富集传代培养过程中菌群结构的变化,保证窖泥生产质量的稳定性,本研究采用高通量测序技术分析了在富集传代培养过程中窖泥产己酸功能菌液的细菌群落结构及物种多样性。Alpha多样性分析显示,未经传代培养的窖泥富集菌液的物种丰富度显著高于3次传代后的产己酸功能菌液;Beta多样性分析结果表明,未经传代培养的富集菌液和传代1次的菌液微生物群落结构组成较为相似,与传代3次后的群落结构具有显著差异;经过多次传代后,产己酸功能菌液菌群趋于相对稳定,狭义梭菌属(Clostridium_sensu_stricto_12)、拉梅尔芽孢杆菌属(Rum-meliibacillus)、Sporanaerobacter、沉积菌属(Sedimentibacter)和普雷沃氏菌属(Prevotella)是优势物种群体。本研究有助于深入了解产己酸功能菌液微生物的多样性组成,可为指导产己酸窖泥功能菌液的扩大培养及工程化应用提供一定的理论依据。In order to ensure the stability of pit mud production,we used high-throughput sequencing technology to analyze the bacterial community structure and species diversity of the caproic acid-producing functional bacterial solution during enrichment and subculture.Alpha diversity analysis showed that the species richness of the uncultured bacterial solution was significantly higher than that of the bacterial solution cultured for three gener-ations.Beta diversity analysis showed that the microbial community structure of the uncultured bacterial solution was similar to that of the bacterial so-lution cultured for one generation,and was significantly different from that of the bacterial solution cultured for three generations.After multiple gen-erations,the bacterial community of the functional bacterial solution was relatively stable,with Clostridium_sensu_stricto_12,Rummeliibacillus,Spor-anaerobacter,Sedimentibacter and Prevotella as the dominant species groups.This study is conducive to the understanding of the microbial diversity composition of the caproic acid-producing functional bacterial solution,and provides a theoretical basis for guiding the expansion culture and engineer-ing application of the caproic acid-producing functional bacterial solution.
