摘 要:通过正交设计法对香菇真菌发酵培养基中氮源最佳配比进行探索,结果表明,豆饼粉为菌体生长的最佳氮源.几种氮源物质的质量最佳配比为:w(豆饼粉)∶w(酵母膏)∶w(蛋白胨)=1∶1.5∶0.2.在此条件下,香菇多糖的产量在发酵130h时最大,其浓度可达25.1mg/mL.该多糖经乙醇沉淀、Sevag法除杂蛋白,用SephadexG-75柱层析分离,得到两种组分的香菇多糖.Orthogonal design was used to explore the optimum organic nitrogen sources for lentinus edodes fermentation and the best ratio of the three nitrogen sources in the medium is that bean powder is 1, yeast extract paste 1.5, tryptone 0.2. The results also indicate that bean powder is the best nitrogen source for lentinus edodes fermentation. Subsequently, lentinus edodes was cultured in the optimum medium for 130 h and the concentration of lentinen in the fermentation medium could reach the climax of 25.1 mg/mL. lentinen was crudely extracted from the liquid medium by alcohol precipitation, then deproteinized by the Sevag method from the polysacchrides and the deproteinized lentinen was purified by gel filtration with Sephadex G75 to obtain two components of lentinen.
