固定化根霉发酵生产脂肪酶

摘　　要：以聚氨酯为少根根霉固定化载体 ,对固定化后的细胞连续重复批次发酵进行了研究。优化了重复批次发酵培养基组成。在取代发酵液 40mL ,取代培养基组成为全脂豆粉 3 % ,花生油 0 5%条件下 ,固定化菌体摇瓶实验可连续使用 14 0h ,重复 9批次。酶的时空产率提高 6倍。 5L发酵罐小试固定化菌体可连续发酵 6批次。固定化细胞连续发酵 ,大大缩短了发酵的时间 。Repeated batch fermentation with Rhizopus arrhizu s immobilized by polyurethane was optimized in details including the time to r eplace, the volume of the replaced medium and the optimal composition of the med ium to replace. Immobilized cells showed stability for repeated use. Immobilized cells could be used for 9 batches, lasting 140h in flask and 6 batches in 5L fe rmenter when the substitute culture medium consisted of (%):soybean flour 3 0 , earthnut oil 0 5 and the liquid was replaced by 40%. The time for fermentatio n was reduced largely and the lipase productivity was 3～6 times as high as that in batch fermen^tation.
