摘  要:从宇佐美曲霉(Aspergillus usamii)E001中克隆木聚糖酶基因xynⅡ,将不含原酶信号肽编码序列的xynⅡ以正确的读码框克隆到大肠杆菌表达载体pET-28a中,构建成重组质粒pET-Xn并转化大肠杆菌,得到重组工程菌KN21。通过对其进行IPTG浓度、温度等诱导培养条件的优化后,胞内重组木聚糖酶活力最高达到81.77U/mg。对重组木聚糖酶进行酶学性质研究,显示其最适温度为45℃,最适pH为4.5。The gene xyn Ⅱ encoding the xylanase was cloned from Aspergillus usamii E001. The xyn Ⅱ without sequence encoding original signal peptide were cloned into the plasmid pET-28a in correct reading frame, then the recombinant plasmids pET-Xn was transformed into the host E. coli, and the recombinant strain KN21 was obtained. After optimization of induction, the expression level of xylanase exceeded 81.77 U/mg finally. The optimum temperature and pH of the recombinant xylanase were 45℃ and 4.5 respectively.
