摘  要:从富含半纤维素的土壤中分离、纯化木聚糖酶的高产细菌,并对其酶学性质进行研究。采用平板法及DNS法分离筛选菌株,利用16SrDNA鉴定菌种;研究不同pH、温度、金属离子等因素对木聚糖酶酶学性质的影响。最终筛选出1株木聚糖酶高产细菌,经鉴定其为类芽孢杆菌。对其酶学性质的研究表明,其最适温度50℃,最适pH5.0;在pH4.0~10.0、温度20~50℃之间有较好的稳定性,其野生型菌株的木聚糖酶活力可达到122.77IU。According to xylan-hydrolyzed halos method and DNS reagent assaying xylanase activity, a high xylanase-producing bacteria was isolated from the soil-samples which full of high hemicellulose, and the enzymetic property of xylanase were studied at the same time. Based on 16S rDNA identification, the novel isolate belong to Paenibacillus. sp coming from the Northeast Forestry University. Its xylanase with an apparent optimum temperature for xylanase activity of 50 ℃ and an apparent optimum pH of 5.0, when the temperatures at between 20 ℃and 50 ℃, and pH between 4.0 and 10.0, the xylanase activities were still very high. Xylanase activity of the wild bacteria can reach the maximal level about 122.77 IU.
